This experiment used the variety of Matsumae which was induced from Japan as recipient material,used Zizania.caduciflora of approaching fringe of rice as the donator,and used the aisle method of pollen tube imported the genome DNA of recipient Zizania.caduciflora.After the cultivation of two generation,the variation of phenotype of T1 generation(the shape of plant and spike,the feature of kernal) was not obvious, but the statistical analysis results indicated that period of duration and effective tillering number have significant deviation.Discovered significant plant of phenotypic variation in the T2 generation,and the type of its offspring was abundant.Compared with contrast control,plant of phenotypic variation had such appearance as period of duration prolonged,plant was flourish,tillow increased and spike was long,the appearance of part of them was that the plant was dwarf,had little tillow and the shape of grain was big,small and long,and the appearance of part of them had awn and their color were violet.Two plants of T2 generation had no ability to procreate.Selected 22 extraction DNA from variants of T2 generation randomly,and did southern blot with the genome of Zizania..caduciflora,the analytic result indicated that the mutation individual of T2 generation really had some plants including of exclusive DNA sequence ofZizania.caduciflora.The transposon was the moving and extendible geno- elementum in the genome.Study the transposition of transposon which was in the condition of definite derivation can help us to understand the biology significance and function of transposition.Did Southernblot and transposon analysis in the condition of choosing phenotypic variation plant randomly,discovered that the mping which pertaining to transposon of MITET category.Devised the specific site primer which including of mping according to the genome sequence of Nipponbare.Detected that T2 generation had the dap of mping by used the method of TAIL-PCR, but the dap was frequent and had no regularity.Studied the transposition activity of retrotransposon TOS17 which had been reported that existing the potentiality of activation at the same time,and the result indicated that the variant of T2 generation can activate the transposition of TOS17.This experiment obtained the excellent variation offspring of rice,especially the early generation of variation,and this enlarged inherited resource and circumsciption of seed selection for breeding worker.The results of this thesis offered the new theory bases for the possible role of distance hybridization in the genome evolution and inheritance improvemen of plant.
