The pollen-tube pathway transformation technique can bypass the usual tissue culture and simplify soybean transformation procedures, but it is restricted in transgenic soybean because no sufficient evidence such as Southern-blot and heredity data to support exogenic gene to integrate into chromosome of soybean.In this paper, pBI121 plasmid is transformed soybean by pollen-tube pathway. 2 transgenic soybean are gained by PCR RT-PCR GUS histochemical assay. Southern-blot. T2 plants of the 2 transgenic plants are analyzed by PCR and positive plants are gained which shows the exogenic gene can inherit generations. The experiment proves transformed soybean can be realized by pollen-tube pathway.vector backbone sequence integrated into plant chromosome is a hotspot in transgenic plants. Recent studies showed vector backbone sequence may affect the expression of natural gene in plants and hasten recombination of transgene.In this paper, the gus gene linear construct lacking vector backbone sequence (only contains promoter, open reading frame, and terminator) are transformed soybean by the pollen-tube pathway. By PCR, RT-PCR, GUS histochemical assay. Southern-blot, 8 positive plants are gained. T2 plants of the positive plants are analyzed by PCR Southern-blot and T3 plants of a positive plant are detected , positive plants are gained which shows the exogenic gene can stably inherit generations. This experiment proves the gus gene linear construct can be transformed into soybean by pollen-tube pathway. So, it will be believed that the use of pollen-tube pathway to deliver the linear gene construct lacking vector backbone sequence into soybean will become a choice for generating transgenic plants in agronomic and industrial interest.It has been concerned that Multi-gene co-transformed into plants to achieve multi-properties' transgenic plants. In this paper, the bar gene linear construct lacking vector backbone sequence is construed. The construct and the gus gene linear construct are co-transforomed soybean by pollen-tube pathway. This worker is prepared for co-transformed in the lab.
