The research on the components of medicinal plant is an important ingredient of the traditional Chinese medicine modernization, of which the study on bioactive carbohydrate is a focal point at present. The present paper described the preliminary investigation on components and fractionation method of monosaccharides and oligosaccharides of ethanol soluble part of water extract from fruit bodies of Bulgaria inquinans (Fries) and extraction process of water-soluble polysaccharides from Epimedium brevicornum Maxim., which could provide great valuable references for further exploitation of the two medicinal materials.Bulgaria inquinans (Fries) is a special local medicinal and edible fungus of Changbai mountain area, which has many pharmacological activities. The study on the components of ethanol soluble part of water extract indicated that it appeared as deep brown powder and contained large amount of monosaccharides, oligosaccharides and steroids small molecule compounds. The monosaccharides were mainly glucose, the oligosaccharides were mainly disaccharides or trisaccharides, and all the carbohydrates were composed of mannose, glucose and galactose in a molar ratio of 0.22:3.82:1.00. The TLC analysis of the fractions of ethanol soluble part of water extract exhibited a convenient and effective fractionated process: at first, a water-eluted fraction, which was consisted by carbohydrates and steroids, and two ethanol-eluted fractions, which were consisted by two steroids respectively, were obtained by using macroporous resin and eluting with water, 30% and 50% ethanol in turn; then, the water-eluted fraction was extracted by saturated solution of n-butanol and water, applied to a silica gel chromatography column, eluted with chloroform-methanol-water (70:30:10, v/v/v, underlayer); subsequently, the obtained fractions were further purified by reverse-phase silica gel chromatography column to generate purified fractionsEpimedium brevicornum Maxim. is a medicinal plant of Berberidaceae. As a traditional Chinese medicine, it is recorded in many medical books and has effects on hypertension, hyperlipemia, dysfunction, hepatitis and aregenerative anemia. The present study on polysaccharide extraction was used dried Epimedium brevicornum Maxim., which was extracted followed the method of water-extration and ethanol-precipitation, and discussed the effect of the different conditions on the yield of polysaccharide. Finally, the optima extracted conditions were obtained: the ratio of material and water was 1:15 (w/v), extracted 1.5 h with boiling water and repeated three times, used three times volume 95% ethanol to precipitated polysaccharide. Following the above conditions, the yield of crude polysaccharide was 6.37%, the protein content was 1.33% and carbohydrate content was 37.9%.
