Epimedium polysaccharide(Epimedium polysaccharide) is one of the main effective components of Epimedium brevicornum Maxim(Epimedium brevicornum Maxim), It has many kinds of biological activity and functions, for example,it can regulate immune function and resist the virus and resist the oxidant and so on. Data show that epimedium polysaccharide is a kind of good immune modulators of livestock and poultry, has high research value and application prospect.This article explores the extraction process and content determination of epimedium polysaccharide as well as its impact on broiler growth performance and immune regulation.1.Create a method for the content determination of epimedium polysaccharide(phenol-sulfuric acid method). And on this basis, by studying the single factors of the ratio of material to water, extraction temperature, extraction time, extraction times and the four factors three levels orthogonal experiments, the extraction process of epimedium polysaccharide was optimized.And get the best extraction conditions: the ratio of material to water in 25, extraction temperature is 90 ℃, extracting time for 2 h, extraction times for 3 times.At the same time, we study the concentration of alcohol precipitation, and put the 70% alcohol concentration as the best concentration of alcohol precipitation.2.According to the optimal extraction conditions to extract epimedium polysaccharide and process the alcohol purification.By using phenol-sulfuric acid method to measure the content of epimedium polysaccharide.and we get the average yield of epimedium polysaccharidewas 5.81%, the average content of polysaccharide of epimedium crude polysaccharide was 42.36%, the average content of epimedium polysaccharide was 2.46%.3.Divide the best epimedium polysaccharide that use the best conditions to extract and process alcohol purification into different doses for animal experiments, The experiment divided into epimedium polysaccharide group, the immune suppression group and blank control group, the immune suppression group used immunosuppressant cyclophosphamide(CY) for immunosuppression, epimedium polysaccharide group included the low dose, middle dose and high dose was studied the production performance and immune function of broilers. The results showed that different doses of epimedium polysaccharide on broilers’ average daily gain(ADG), feed conversion ratio(FCR) of production performance has different degrees of increase; Different doses of epimedium polysaccharide on the immune organ index, immunoglobulin, cytokines has enhanced role; In different experiment time, the immune index of the epimedium polysaccharide experimental group was significantly better than the immune suppression group through the statistical analysis(P<0.05), and middle dose or high dose of some immune index better than the blank control group, but has yet to reach significant difference(P>0.05).4.Divide the best epimedium polysaccharide that use the best conditions to extract and process alcohol purification into different doses for animal experiments, The experiment divided into epimedium polysaccharide group, the immune suppression group and blank control group, the immune suppression group used immunosuppressant cyclophosphamide(CY) for immunosuppression, epimedium polysaccharide group included the low dose, middle dose and high dose was studied for the influence of the intestinal morphology and anti-oxidation function of broilers, the results showed that different doses of epimedium polysaccharide can increase intestinal villus length of broilers in different degrees, lower recessus depth in different degrees, increase the number of some intestinal beneficial bacteria such as lactobacillus, reduce the number of intestinal harmful bacteria such as escherichia coli,salmonella;Different doses of epimedium polysaccharide can enhance the activity of antioxidant compounds SOD, GSH-Px, CAT in different degrees and reduce the activity of oxide material MDA in different degrees. And in different stages of the experiment, different doses of epimedium polysaccharides was compared with immunosuppressive group, has significant difference in experimental index(P<0.05), and some detection index better than the blank control group, but has yet to reach significant difference(P>0.05).
