Lentinus edodes (L. edodes)is a well known macrofungi with edible and medicinal value, its polysaccharides have a variety of biological and immunological activity, such as anti-tumor effects, stimulate the formation of interferon, strengthen host resistance to bacterial, fungal, viral or parasitic infections, enhance natural killer (NK), cytotoxic T lympjocutes (CTL) activity and so on.Its chemical structure is great osculation with its activity,and so,L. Edodes's chemical structure is gradually investigative hotspot.By this article,we took the dry mushroom L. Edodes for experiment material,and research the structure of the low molecular weight polysaccharides which can soluble in water.It was lay the foundation for low molecular weight polysaccharides's studies on locomotor activity.In this paper:dry mushroom L. edodes were extracted with distilled water for three times, then the supernatant was concentrated and precipitated with ethanol to obtain the crude polysaccharide (designated C-LNT-1). C-LNT-2 was recovered by repeated frozen, thawed and precipitating with 95% ethanol from C-LNT-1. C-LNT-2 was deproteinated by a combination of proteinase and Sevag method and was further purified by Sepharose CL-6B gel filtration chromatography to get the homogeneity and low molecular weight lentinan(Mw=7040Da)(designated LMw-LNT)which was evaluated by HPLC. Gas chromatography (GC) analysis showed that LMw-LNT was composed of Glc, Gal, Man, Ara and Fuc in ratio of 10.74:4.78:2.3:1:1.04.We can knew the LMw-LNT 's molecule structure by partially hydrolyzed by some method of structural research.It has a backbone mainly composed of(1→3)-linked Glc,and (1→6)-linked Gal and a little (1→2)-linked Fuc with 4 branches attached to (1→3,5)-linked Ara, (1→2,4)-linked Man and terminated with (1→)-linked Man,and the(1→4)-linked Gal,other (1→3)-linked Glc,a little (1→2)-linked Fuc and (1→3,5)-linked Ara are distributed in branches.
