QTL Mapping For Agronomic Traits In Chromosome Segment Substitution Lines Of Rice

Using CSSLs derived from Sasanishiki /Habataki cross, Koshihikari/Kasalath cross as material, this research selected some heterozygous locus originated from CSSLs derived from Sasanishiki/Habataki cross and expanded three lines which overlapped the chromosome 1 of CSSLs constructed by Koshihikari/Kasalath. At the same time using the CSSLs derived from Sasanishiki/Habataki cross some agronomic traits such as plant height, panicle length, seed-setting rates and protein content were analyzed. In 2007 and 2008, these materials were planted twice at Yangzhou, one time at Hainan. Then the relationship between QTL and environment was analyzed and correlated QTLs were located. The results were as follows:1 Four pure lines were got through selecting prior lines with heterozygous locus originated from the CSSLs derived from Sasanishiki/Habataki cross by using the SSR markers, which build good foundation for the QTL location.2 Three lines which overlapped chromosome 1 of CSSLs derived from Koshihikari/Kasalath cross were expanded. Then 7 new lines were got, the original fragments were reduced, and the precision of QTL location was enhanced.3 Using the CSSLs derived from Sasanishiki/Habataki cross, 17 plant height QTLs were located, and three of them were detected three times; two of them were detected twice. At the same time 13 panicle length QTLs were located, one of them was detected three times, and another two QTLs were detected twice respectively.4 Using the CSSLs derived from Sasanishiki/Habataki cross, 9 Seed-setting rates QTLs were located and the QTL qSSR-12-1 on chromosome 12 was detected three times, and it showed greatly effect value. By detecting the CSSLs'panicle fertility and pollen fertility of the QTL qSSR-12-1, and making the electro microscopy scan of the embryo slice, the reason of sterility was validated as the male gamete abortion. The genetic analysis showed that this loci was according with lethal mode of duplicated recessive gamete.5 Using the CSSLs derived from Sasanishiki/Habataki cross, 13 protein content QTLs were located. These QTLs qPC-1-1, qPC-1-2, qPC-1-3, qPC-3-1, qPC-5-1, qPC-10-1 were detected twice. qPC-1-2 and qPC-5-1 showed different additive effective at Yangzhou and Hainan respectively, it suggested the protein content was immensely influenced by the environment.
