PRL which secreted from animal anterior pituitary gland in eosinophils is the peptide hormones by a single gene encoding.It exists in almost all vertebrates.It is an animal lactation,egg production and other functions necessary for reproduction,a wide range of biological effects.At present,PRL gene(prl) has been considered to be associated with poultry breeding performance of the candidate genes.Sichuan-White geese are of the fine species placed the "varieties of Chinese poultry"(1987), laying performance with good,quick growth,wide adaptability,good effects,such as hybrid varieties of unique characteristics of the merchandise in our country goose production occupies a very important position.Reproductive traits are used to measure the level of poultry production performance of one of the most important economic traits.However,because of poultry reproductive traits are affected by many minor genes control quantitative traits,influenced by genetic,nutritional, environmental,disease and many other factors,low heritability.The conventional breeding program has resulted in the production performance of poultry almost its limit.With the rapid development of biotechnology,molecular biology used to study the use of reproductive traits of the major gene,a candidate gene and the search for genetic markers has become an effective way to improve poultry breeding fertility research.In this study,130 genome DNA of Sichuan-White geese blood samples for prl exon 2 and 5 were SNP analyzed by PCR-SSCP technology.To obtain these results,not only for the Sichuan-White goose prl function studies provide a reference,but also to its polymorphism provide the basic data.In addition,the test using least squares analysis of the prl exon 2,5 flocks of different genotypes had been related to egg production analysis,expected the discovery of new fecundity and Sichuan-White goose related molecular markers.The contents and results of this study are as follows:1 SNP analysis of Sichuan-White geese prl exon 2 and 5Selected 130 healthy adult female Sichuan-White geese respectively collected their tibial vein blood and listed under the wing markings.Extracted blood samples of 130 Sichuan-White geese genome DNA,as a template for its prl exon 2 and 5 of the PCR amplification.PCR products by 12% polyacrylamide gel electrophoresis analyzed and silver stained.PRL gene exon 2 was detected having AA AB two genotypes in Sichuan-White geese;their frequencies are 0.6231 and 0.3769.A allele frequency is 0.8115,B allele frequency is 0.1885.There also has CC CD two genotypes at exon 5,their frequencies are 0.6769 and 0.3231.C allele frequency is 0.8385,D allele frequency is 0.1615.These different genotypes cloning and sequencing of the PCR products were found,there is a SNP loci,that is,the A/G mutation at 141 causes the replacement of amino acids from Arg to Gly at exon 2 of Sichuan-White goose PRL gene.Exon 5 is also the existence of a SNP loci,that is,the T/C mutation at 135 causes the replacement of amino acids from Leu to Pro.2 The relationship between Sichuan-White geese prl exon2,5 and laying performanceIn accordance with detected genotypes AA AB CC CD at exon 2 and 5 of PRL gene, respectively 79 Sichuan-White geese of the laying period were partitioned groups for egg production. Single factor least squares linear model analysis results showed that egg production of four genotypes no significant differences(P>0.05).However,homozygous genotype is greater than the least squares means for genotype heterozygosity,that is,AA>AB,CC>CD.
