Fibroblast growth factor 5 (Fibroblast Growth Factor-5 FGF5) is a cyclical growth of skin hair follicles important control factor. Using RT-PCR method has been proved that the FGF5 gene in cashmere goat skin follicles cyclical period of growth and prosperity in the hair follicle catagen are detected mRNA expression; this study by observing the different stages of Inner Mongolia Cashmere goat skin biopsy, the use of PCR technology fragment was amplified, cloned gene fragments and the design of probe, and finally through the in situ hybridization detection of the gene in the skin-targeted expression of the organization. The results showed that: in situ hybridization from the point of view of the picture FGF5 gene in cashmere goat skin hair follicle growth in the hair follicle cyclical prosperous period, catagen and telogen are targeted expression.The use of PCR-SSCP method of the same age in Inner Mongolia White Cashmere goats FGF5 gene analysis of single nucleotide polymorphisms. The results showed that: FGF5 gene in P2 fragment amplified in PCR-SSCP polymorphism exists. PCR product by sequencing, the amplified fragment of the first 387 points, the existence of C→T mutation, but there is no change of amino acid for glycine. The primer also has 301 points on the T insertion phenomenon, caused changes in amino acid from the CCT proline→TCC serine. Individual goats of different genotypes and gene frequencies of the results show that the primers in the Inner Mongolia White Cashmere goats are generally based allele A, allele frequency in the Hardy-Weinberg imbalance. Plush velvet of different individuals with other genotypes long correlation analysis showed that very significant differences to velvet and no signigicant differences to plush.Cats on the basis of the 182bp and 194bp found two mutations designed a pair of primers using PCR-SSCP method of cashmere goats whether polymorphism, results showed that: Inner Mongolia White Cashmere goats in the two FGF5 gene locus did not find polymorphism.
