This study is divided into five parts. Firstly, It were analyzed and evaluated that amino acid profiles in the muscle, hair and ruminal microbial protein in Aohan Merino Sheep. Three target amino acid patterns were established according weight of the amino acid pattern between muscle and hair. It were also calculated that the amino acid flow in the duodenum and the digestibility of amino acids in the small intestine of Aohan Fine Wool Sheep fed basal diet in vivo. Then doses of amino acid infusion for different patterns were calculated according to the Amino Acid Dynamic Addition Model. Twelve amino acids mixture in same amount but with different profile of amino acids were introduodenally infused in order to make ultimately target pattern for absorbable amino acid in the small intestine. amino acid digestion,blood biochemical indicators and performance indicators with different pattern were investigated, so as to find an ideal absorbable amino acid pattern. The results showed that there was an ideal amino acid pattern Conducived to the growth of wool in the intestine of Aohan Fine Wool Sheep . It was suggested that hair performance could improved by regulating absorbable amino acid pattern in the small intestine.Part 1 A Study on target pattern of absorbable amino acids in the small intestine of Aohan Fine Wool Sheep.Nine wethers of Aohan Fine Wool Sheep (35-45kg) with the same father were used to analyze and evaluate the profile of amino acids in the muscle, hair and rumenal microbe protein. The results showed the amino acid profiles were different among muscle, hair and rumenal microbe protein.The contents of Serine, sulfur amino acid, phenylalanine, arginine were higher in the hair,but the contents of Glycine, alanine, lysine, histidine and tryptophan were higher in the muscle.It indicated that there was a clear distinction between amino acids required for wool growth and muscle growth,and Wool growth demanded more sulfur amino acids and serine. Rumen microbial amino acid composition and amino acid composition of wool and muscle were also significantly different. The contents of Cystine, serine and arginine were significantly lower than the levels of wool.With the General analysis of the profile of amino acids in the muscle, hair and rumenal microbe protein, Both the amino acid composition of feed or the rumen microbial protein were not conducive to the growth of wool fiber. So three target amino acid patterns were established according weight of the amino acid patterns between muscle and hair, which were M100,M85+H15 and M70+H30, so as to find an ideal absorbable amino acid pattern in the small intestine for hair producing.Part 2 Study on amino acid flow and digestion in the small intestine of Aohan Fine Wool Sheep fed basal diet.Nine wethers of Aohan Fine Wool Sheep (35-45kg) with the same father, fitted with permanent cannulas in the rumen, at the proximal duodenum and terminal ileum were used to investigate amino acid flow and digestibility by Continuous perfusion technique in different sites of the digestive tract of Aohan Fine Wool Sheep fed the basal diet. The test diet of the same weight were fed daily at intervals of 2h. The results showed the duodenal flow of Sulfur amino acid was smaller and accounted for 2.79% of the total flow of amino acids. The disappearance of Sulfur amino acid in the small intestine only accounted for 2.81% of the total disappearance of amino acid,which was incompatible with the truth that wool production needed Substantial sulfur amino acids and lead to the restrictions on wool production performance of Aohan Fine Wool Sheep. Because of continuous feeding, the digestibility of amino acids were relatively high,and the average digestibility of amino acids in the Small intestinal was 80.45%.The high intestinal digestibility of Met indicated the ability of using sulfur amino acids was high and were consistent with Aohan Fine Wool Sheep as a high-performance sheep.Part 3 Effect of absorbable amino acid levels in the small intestine on digestion of AA in Aohan Fine Wool Sheep.Nine wethers of Aohan Fine Wool Sheep (35-45kg) with the same father, fitted with permanent cannulas in the rumen, at the proximal duodenum and terminal ileum were used to investigate the effect of absorbable amino acid levels with the same ideal pattern in the small intestine on digestion of amino acid in Aohan Fine Wool Sheep. The test diet of the same weight were fed daily at intervals of 2h. Twelve amino acids mixtures in same amount but with different profiles of amino acids were introduodenally infused in order to make ultimately target pattern for absorbable amino acids in the small intestine, which were M100,M85+H15 and M70+H30. Amino acid flow and digestibility in different sites of the digestive tract and the concentration of free amino acid in plasma were researched in three absorbable amino acid pattern. The results showed the duodenal amino acid flow was lower than the flow of the basic diet after amino acids infusion.It indicated that the infusion of amino acid balanced the amino acids profiles in the small intestine and the absorption and utilization of amino acids in Intestinal was enhanced. Apparent digestibility in SI and apparent digestibility in GIT were lower than those of basic diet,and Amino acid excretion in feces were higher than basic diet,which was because excessive amount of amino acid infusion lead to the inadequate digestion and absorption,so we should research the absorbable amino acid level in the small intestine with the ideal amino acid pattern. In three absorbable amino acid pattern, the amino acid flow in Small intestine and feces were lower than the other two groups and the ratio of Gly/OAA of plasma free amino acid was the lowest ,but there were significant amino acid stranded in jugular vein. Overall, the amino acids digestion and utilization of M70+H30 were better than the other two groups,but amino acids had been stranded in blood,so the ideal absorbable amino acid pattern in the small intestine of Aohan Fine Wool Sheep was no more than M70+H30.Part 4 Effect of absorbable amino acid pattern in the small intestine on plasma biochemical index and hormones level in Aohan Fine Wool SheepNine wethers of Aohan Fine Wool Sheep (35-45kg) with the same father, fitted with permanent cannulas in the rumen, at the proximal duodenum and terminal ileum were used to investigate the effect of absorbable amino acid pattern in the small intestine on plasma biochemical index and hormones level in Aohan Fine Wool Sheep. The test diet of the same weight were fed daily at intervals of 2h. Twelve amino acids mixtures in same amount but with different profiles of amino acids were introduodenally infused in order to make ultimately target pattern for absorbable amino acids in the small intestine, which were M100,M85+H15 and M70+H30. Alanine aminotransferase (ALT), plasma urea nitrogen (PUN), growth hormone (GH), insulin (Ins) and IGF-I were determination in plasma. The results showed ALT concentration increased and PUN concentrations decline with the increase in the proportion of the wool pattern. The body protein metabolism was strengthening with the increase in ALT,and the lack of restrictive amino acid or amino acid imbalance in Sheep would improve PUN concentration,so the amino acid utilization of M70+H30 was better than the other two groups. Part 5 Effect of absorbable amino acid pattern in small intestine on the capability of wool production of Aohan Fine Wool SheepNine wethers of Aohan Fine Wool Sheep (35-45kg) with the same father, fitted with permanent cannulas in the rumen, at the proximal duodenum and terminal ileum were used to investigate the effect of absorbable amino acid pattern in small intestine on the capability of wool production of Aohan Fine Wool Sheep. The test diet of the same weight were fed daily at intervals of 2h. The wool growth rate and fineness at 28 days were measured in basic diet or three kinds of amino acid infusion mode. The wool growth rate of M70 + H30 was 3.5 times of the basic diet,so the growth rate of wool significantly increased with the duodenal amino acid infusion. Amino acid balance of the small intestine had also reduced the wool fineness. With the general analysis of wool production, plasma free amino acid concentration, blood biochemical parameters,we obtained the ideal Pattern of absorbable amino acids in the Small Intestine of Aohan Fine Wool Sheep was to get close M70+H30.
