In this paper,'Snow Queen' and 'Siberia' were used to study the compatibility with other lilies.Four pollinations methods,such as cut-style,in vitro pollination,graft-style, and graft-plant were used.During self-pollination and cross-pollination of 'Snow Queen', the development of the pollen growth under fluorescence microscope was observed,and the dynamic changes of protective enzymes and endogenous hormones in pistil were further studied.Meanwhile,the microspore mother cell meiosis abnormality of 'Siberia' was observed.This paper provided the theoretical foundation and the ways for the mechanism of pre-fertilization barrier and hybridization breeding in Lilium.The main results are as follows:1.The compatibility between Lilium pumilum,L.dauricum and 'Snow Queen' was better than the others,and the hybrids were obtained through cut-style pollination and ovary culture.Among the three pollination ways(cut-stylar pollination,normal pollination and cut-stigma pollination),the hybrids were only obtained through cut-stylar pollination. These results showed that the pre-fertilization barrier both existed in the tissue of stigma and stylar.The stigma exudates of 'Snow Queen' were beneficial to the development of pollen tube and more hybrids.And the pre-fertilization barrier was effectively overcomed through cut-stylar with stigma exudates and ovary culture.2.The co-changes of protective enzymes and endogenous hormones were caused by pollination compatibility,which varied with pollen development.The high level of SOD and POD and the contents of IAA,GA3 and ZR were beneficial to compatible pollen adhesion and germination(1-3 h after pollination),and the activities of SOD and POD had a similiar tendency with the contents of IAA,GA3 and ZR during this stage.The content of ABA of cross-pollination was higher than that in self-pollination,indicating that the high level of ABA was related with cross-incompatibility.3.The pollen tube of self- and cross-hybridation stoped in the tissue of stigma,while the pollen was not germinated in the stigma with cut-stylar and cut-stigma.The results showed that pre-fertilization may exsit in the stigma,where was also the prerequisite for pollen germination.Some abmormalities of meiosis of pollen mother cells were studied in Lilium 'Siberia';they were homologous chromosome synapsis abmormalities, chromosome brige,laggard chromosomes,chromosome division asynchrony,micronuclei and so on.The results showed that there were abmormalities in the reproductive system, which may lead to pre-fertilization in 'Siberia'.
