The research of fish as an experimental animal is lag in China,and the standardized animal of experimental fish is still blankness.Goldfish(Carassius auratus Var),which origin provenance is China,could be a experiment animal of fish for study and exploration.The anatomy properties,protein, isodynamic enzyme and randomly amplified polymorphic genome DNA of the goldfish have been studied and reported.Howerver, as a experimental animal,the relationship of system evolution and the inherit molecular background are still arguably.MtDNA is a widely used molecular marker to investigate the phylogenesis of species.In present study,mitochondrial tRNA-Thr gene,tRNA-Phe gene and partial control region(D-loop) and Cyt b of different varieties of goldfishes(Carssius auratus var) and crucian carps(Carssius auratus) had been sequenced.The relationship of mtDNA molecular genetic between these fishes and other related fishes were illuminated.The major results in this paper were presented as follows:1,Mitochondrial tRNA-Thr gene,tRNA-Phe gene and partial control region of representative varieties of Carssius auratus var.,red dragon goldfish,red cap goldfish,tiger head goldfish, gold plating goldfish,black dragon goldfish,water vesicle goldfish and Carssius auratus of Peng Ze were sequenced.The molecular phylogenetic tree of tRNA-Thr gene,tRNA-Phe gene and partial control region sequences among these goldfish,red crucian carp,Japanese crucian carp,and common carp from GenBank was constructed with DNAstar software,tRNA-Thr gene and tRNA-Phe in those different varieties of goldfishes and crucian carps were identical which showed their conservative. Thus the tRNA-Thr gene and tRNA-Phe gene sequences couldn't be a good molecular marker.The control region has important value for investigating intraspecific genetic differentiation because of its high mutation accumulation.The analysis of D-loop showed that the origin of goldfish was crucian carp and the genetic material take a differentiation in the long evolutional process.2,Mitochondrial Cyt b gene of representative varieties of Carssius auratus var.,red dragon goldfish,red cap goldfish, tiger head goldfish,gold plating goldfish,black dragon goldfish,water vesicle goldfish,genuine pearl goldfish and Carssius auratus of Peng Ze were sequenced.The molecular phylogenetic tree of Cyt b sequences among these goldfish,red crucian carp,Japanese crucian carp,silver crucian carp,wild curcian carp and common carp from GenBank was constructed with DNAstar software.The study presented the molecular evidence that the origin of goldfish was from wild crucian.
