We have undertaken comparative studies on blood of red crucian (RCC),blunt snout bream(BSB) and their hybrids including triploids, tetraploids,and pentaploids.The main results and conclusions were as follows:1.The average nuclear volume of erythrocytes in peripheral blood increases regularly with the increasing of ploidy.Hence,the mean size of erythrocytes nuclear could be used to identify the ploidy as one of the criterions.Furthermore,many more mature erythrocytes have a dumbbell nucleus in the peripheral btood and hemopoietic organs of polyploidy hybrids compared with their diploid parents.With the increase of ploidy level,the percentage of such erythrocytes increases.The polyploidy hybrids also have a large number of erythrocytes with abnormal shapes. For example,round and tear-shaped erythrocytes have been observed in the peripheral blood of polyploidy hybrids.The morphological variation of mature erythrocytes in polyploidy hybrids could be used as a useful marker to identify polyploidy hybrids from diploid.2.Primitive hematopoietic cells of RCC,BSB and their polyploidy hybrids were observed and compared.The results indicated that the spleen and kidney of polyploidy hybrids also had the ability of hemopoiesis.The number of primitive hematopoietic cells decreased with the enlarging size of cells in hemopoietic organs of polyploidy fishes, suggesting that the number of cells in 3nRB,4nRB,and 5nRB hybrids deceased to keep their hemopoietic organs in normal size.3.Protein electrophoresis was utilized to analyze and compare the peripheral blood hemoglobin and total protein of RCC,BSB and their polyploidy hybrids.The results demonstrated that there was no apparent difference of the hemoglobin subunit about the above fishes.But there were some differences of total protein in different ploidy fishes.4.The sequences of cdc2 partial cDNA in RCC,BSB and their polyploidy hybrids were obtained and these sequences were compared and analyzed.The similarity about these sequences was over 98.2%.It implied that the sequences of cdc2 in these fishes were highly conserved. The results of SqRT-PCR demonstrated that transcriptional level was increased with the increase of ploidy.This phenomenon may relate with the function of p34cdc2 in condensing chromosome.Also it may be relevant to the high condensation of erythrocyte nuclei in polyploidy and the tendency of prolification.
