In the modern agricultural production, continuous cropping obstacle is always one big problem to us. Many ways were attempted, but there were no effective methods to overcome continuous cropping obstacles. This thesis was to discover an effective method of overcoming continuous cropping obstacles of watermelon by using four different microbial amendments on continuous cropping soil. And soil microbial before and after using microbial amendments on continuous cropping obstacles soil was determined by using DGGE. Besides the growth, the qualities and the yields of watermelon were determined. The crop rotation experiments of fruit vegetable and leaf vegetable were made in the same time, soil microbial before and after rotation were determined by using DGGE. Thinking about the problem of cost, choice of parameters of microbial amendments was done also.The results of DGGE showed that rotations had some effect on soil microbial of continuous crop obstacle, increasing density of soil microbial, changing structure of soil microbial, but the effects were limited.Results showed that four kind microbial amendments can inhibit continuous cropping obstacle obviously. Not only can it improve growth of plant, increase yields, but also improve qualities of watermelon. Analysis using DGGE indicate four kind microbial amendments can improve soil micro flora, increase contains of beneficial bacteria of soil. Using software Quantity one analyzed DGGE map, we find that the treatment of microbial amendments was better than rotation.The result of sequencings demonstrate the primary bacteria affected by each treatment were uncultured bacteria and Proteobacteria and alpha , gamma of Proteobacteria, some were Acidobacteria, Microbacterium and Actinobacteria.Conclusions indicated that using 7g of Lianchawang per plant was best for cost and yields, using 5g of Hgenbao per plant was best for cost and yields, using 1:1000 of concentration of Zhibaokang had maximum economic benefit, using 1:250 of concentration of Shengtiao had maximum economic benefit.
