Cordyceps sinensis, a classical and valuable traditional Chinese medicine what have been favored, have distinct pharmacological actions because of containing especial chemical substances. The culture of C. sinensis become one of important topics to solve urgently due to the lack of the natural resources. The process of the culture, the preparation of C. sinensis has become a important elment which can develop the culture of C. sinensis. In this study, based on these reasons, the artwork prescription of oil suspension formulation was discussed, the condition of solid fermentation wan optimized and the dynamics model of solid fermentation was established, thus making solid basis for the further research on the praperation of C. sinensis and the production of conidiospores by solid fermentation.During the cultivation of C. sinensis, the infection rate of Hirsutella. sinensis is always one of the key factors of limiting its commercialization. In this experiment, according to the particularities of oil suspension formulation can raise the infection rate of H. sinensis to the host, we screened the optimal combination project of oil miscible flowable concentrate of H. sinensis by orthogonal design, range analysis of MATLAB software and the principle of comprehensive balance, namely, a mixture of rapeseed oil and dibutyl phthalate (7: 3) 10 g, K12 0.9 g, fatty alcohol polyoxyethylene ether 0.5 g, aluminium stearate 0.6 g, solid-liquid ratio 1.0: 10. As a result, the preparation's sedimentation volume ratio was 97% and redispersibility 100%. The results of stable test on the oil suspension indicate that the preparation has favourable stability for storage lasted for 1-2 month.Basis on the research of the oil suspension of C.sinensis, it is important to improve the activity of the oil suspension. According to the characteristics of the oil suspension formulation, MTT assay is adapted and optimized. The result is as follows: reaction time 120 min, reaction temperature 37℃, methylbenzene as extracting agent, and a positive linear correlation established between active cell weights and cell viability.Varieties and concentrations of assistance agents in oil suspension formulation have been selected with the refined MTT assay, and further optimized together with cell concentrations through orthogonal experiment. The optimal combination project was obtained, namely, cell concentration 0.15 g/mL, aluminium stearate 60 mg/mL, and SPAN-80 50μL/mL. The results of the experiment on cellular viability indicate, to the pure cellular viability, the activity of the oil suspension can occupy at the range of the 95%-89%. The results of stable test on the oil suspension formulation indicate that cell viability can maintain above 90% at 4℃after one month.The solid fermentation of C. sinensis was studied. The contrast experiment results showed that the adequate carbon source is glucose and the adequate nitrogen source is yeast extract. At the same time, the dynamics model of the solid fermentation which C. sinensis was cultured on is established through the method of the nonlinear least square curve fitting on the MATLAB soft, thus making solid basis for prehension the essence of solid fermentation, the further enlargement of the solid fermentation, the optimization of the process control and the increasing yield.
