As we all know, Chinese caterpillar fungus are rare medicinal crops in our country, whileCordyceps militaris is the first one of Chinese caterpillar fungus of fungi which can fullyrealized artificial cultivation.One of the effective ways to solve the problem of the scarcityof caterpillar fungus resources is Large-scale cultivation of those.We use three strains of Cordyceps militaris preserved in our lab to study the artificialcultivation and effective composition analysis in this experiment with following work:1. With the index of formation of Cordyceps militaris, liquid and solid fermentationcapability, we screening in the three strains of Cordyceps militaris, then we find that C3is themost appropriate strain in our experiment with its robust hyphae and excellent fermentability no matter in liquid or in solid.2. Previous experiments have found that the strains of Cordyceps militaris degenerateeasily with following phenomenon: increased Liquid fermentation by-products, extendedsolid fermentation period and without entity in Solid fermentation and so on. Withconventional preservation method in this experiment, we find that the best medium forpreservation is PDA medium and it should be preserved at4℃,the time should not be morethan four months and it should be activated after4months, the passage is appropriate withinfour generations in activation.3.Through a single factor and orthogonal experiments of Cordyceps militarisfermentation medium and fermentation conditions to obtain the best Cordyceps militarisfermentation medium which is: sucrose2.0%, peptone0.8%,0.5%KH2PO4, MgSO40.25%.Optimum fermentation conditions is: seed age120h (five days), inoculums’ size10%, thefermentation broth of initial pH6.8, liquid volume200ml/500ml, rotation speed130r/min,the incubation temperature was24℃; optimal fermentation medium and fermentationconditions, the maximum mycelia biomass13.9g/L, the polysaccharide amount46.8mg/g.4.Through the single factor experiment concerning carbon source、nitrogen source andratio of solid to liquid of fermentation of Cordyceps militaris for fruit body, it concludes thatthe optimal carbon source is wheat, the optimal nitrogen source is silkworm pupa powder, andthe optimal ratio of solid to liquid is1:1.2; To optimize the light and temperature of fruitbody of Cordyceps militaris during the period of weed growth, the result is that the propertemperature for the growth of fruit body is24degrees. Light is the necessary condition for thegrowth of fruit body of Cordyceps militaris and the time under light is beyond16hours every day by the experiment.5. Optimize the extracting technology of the cordycepin of fruit body of Cordycepsmilitaris under artificial culture. Select the capillary electrophoresis as detection method ofcordycepin. Through the single factor experiment and orthogonal design regarding extractionliquid、ratio of solid to liquid、extraction temperature and extraction time,it can be concludedthat the optimal extraction liquid of cordycepin is60%alcohol,ratio of solid to liquid is1:20,extraction temperature is70degrees,extraction time is9hours.
