In this study,cotyledonary nodes in Lespedeza bicolor were used for transformation,firstly,factors that affecting regeneration systerm including cultivation conditions and plant growth regulators,etc,were also investigated in details.On this base,regeneration systerm was optimized.Subsequently the Sac B gene was introduced into L.bicolor by Agrobacterium tumefaciens-mediate and the resistant plants were obtained.The expression of the Sac B genes in transgenic plants was confirmed by PCR,PCR-Southern and Southern Blotting,Finally,salt-tolerance test indicated that transgenic plants could grow well in the mediun with 140mmol/L NaCl or 4%PEG.The present study may be the basis for obtaining a more drought-resistant L.bicolor.
