Reproductive organs of female mammal are known to undergo dynamic cyclic changes during the oestrus cycle.Apoptosis plays an important role in this change.In the present study, in situ fluorescent labeling(DAPI)and immunohistochemistry were used to confirm and locate apoptosis cells and to detect the expression of Bad in major reproductive organs(ovaries, oviduct and uterus)of Small-tails Han Sheep(SHS)and Suffolk Sheep(SS),which have high and low productivity respectively.The purpose of this study was to investigate the apoptosis mechanism of cyclic change of ewe reproductive organs and provide theory for explaining the influence of apoptosis to productivity of ewes.The finding of in situ fluorescent labeling(DAPI)indicate that apoptosis might take place in follicular epithelium cells(FeC),granule cells(GrC),theca cells(ThC)and luteal cells(LuC) of SHS and SS.SS had more apoptosis of GrC and ThC in tertiary follicle and graafian follicle (VG)than SHS.Very few secretory cells(SeC)with apoptosis feature were detected in all segment of oviduct epithelium of SS during luteal phase(LP),while the same phenomenon was not found in SHS.Minority apoptotic cells were detected in subcutaneous connective tissue. Neither SHS nor SS apoptotic cells were detected in oviduct during follicular phase(FP).A great quantity of apoptotic cells were detected in stroma and glandular organ of endometrial during LP,and glandular organ of endometrial had no apoptosisi cells in SHS and SS,while a few endometrial stroma cells(StC)with apoptosis were detected.The finding of Bad immunolocalisation indicated that:Expression of Bad in reproductive organs of SHS and SS in a regular pattern,and submitted location-and cycle-dependence.The expression regularity of Bad in reproductive organs of SHS and SS was the same in general: 1.Average optical(AO)of oocyte in primordial follicle,primary follicle and secondary follicle was higher significantly than in tertiary follicle(P<0.01);2.The ratio of positive Follicular epithelium cells and AO of secondary follicle was higher significantly than primordial follicular and primary follicle epithelium cells;3.The ratio of positive luteal cells and AO of corpus luteum in anaphase was higher significantly than in prophase(P<0.01);4.The ratio of positive cells and AO of uterine tubal epithelium in LP was higher significantly than in FP(P<0.01).5. AO of endometrial gland in LP was higher significantly than in FP(P<0.01);6.The ratio of positive cells and AO of endometrial stroma in LP was higher significantly than in FP(P<0.01).The expression of Bad had several visible differences in reproductive organs of SHS and SS:1.The ratio of positive GrC in tertiary follicle of SS was higher significantly than SHS (P<0.01);2.The ratio of positive cells and AO of ThC in tertiary follicle and vesicula graafianae of SS was higher significantly than SHS(P<0.01);3.AO of uterine tubal epithelium cells had significant deviation between SHS and SS(P<0.01),but there was not regularity to abide.4.AO of Endometrial gland of SS was higher significantly than SHS in FP(P<0.01);5.The ratio of positive cells and AO of endometrium cotyledon stroma of SHS was higher significantly than SS in LP(P<0.01).It is concluded from the results mentioned above that apoptosis is important regulatory mechanism of cyclic change in reproductive organs of ewe,apoptosis gene Bad participate in regulation of this cyclic change.The difference of apoptosis in reproductive organs and expression of Bad between SHS and SS maybe is groundwork which result in differential productivity of their.
