The thesis used the 4×5 NCII design for the hybrid combination material,measuresed the heterosis of the soybean agronomic and quality traits,analyzed the relationship between differential gene expression patterns,proportion and heterosis.In this foundation,we selects an obvious superiority hybrid combination (17×6) according to the fat.Eighteen differentially expressed of cDNA fragments between hybrids and their parents were cloned,and then carried through homologous comparea and presumed function in thses fragments.Expectation that will offer valuable information on the relationship between differential gene expression patterns and heterosis in hybrids and their parents of soybean.The main result as follows:Computed the mid-parent heteriosis(MPH)and the better parents heterosis(BPH) of the soybean different combination.Discover the soybean exists an obvious heteriosis,the average MPH of each trait in the twenty combinations respectively is:plant height is 0.26%,No.of nodes per stem is 0.61%,No.of branches per plant is 20.60%,No.of pods per plant is 28.78%,No.of seeds per plant is 32.72%,insect infected grain is-15.68%,stem diameter is 12.99%,100-seed weight is 7.75%,yield per plant is 43.90%,oil is 1.02%,protein is-0.11%. Among them,most of these combinations,the heteriosis of stem diameter,100-seed weight and the yield per plant is positive advantage,but plant height,No.of branches per plant and insect infected grain is negative advantage,none of traits is positive advantage or negative advantage in the twenty different combinations. Among the traits,the yield per plant is the biggest advantage:the MPH of the combination(3×12) is 139.73%. The average BPH of each trait in twenty combinations respectively is:plant height is-7.44%,No.of nodes per stem is-4.14%,No.of branches per plant is-4.55%,No.of pods per plant is 14.67%,No.of seeds per plant is 17.00%,insect infected grain is 15.14%,stem diameter is 7.28%,100-seed weight is 3.42%,yield per plant is 25.40%,oil is-3.64%,protein is-2.98%.Among them,most of these combinations,the BPH of 100-seed weight and the yield per plant is positive advantage,but plant height,oil and protein is negative advantage, among the traits,the yield per plant is the biggest advantage:the BPH of the combination(3×13)is 117.45%.We also analyzed the combining ability of the agronomic and quality traits on the twenty combinations. To the general combining ability(GCA),only the plant height,100-seed weight and the protein that of the parents all come to 0.1 remarkability.To the female,yiedld per plant and the No.of seeds per plant come to 0.01 remarkability,No.of pods per plant,stem diameter and 100-seed weight come to 0.05 remarkability,oil and protein come to 0.10 remarkability;but at the male parent,the plant height,the No.of nodes per stem and the protein come to 0.01 remarkability,100-seed weight come to 0.05 remarkability,No.of branches per plant come to 0.10 remarkability.To special combining ability(SCA),only the oil come to 0.01 remarkability,other triats are unremarkable.we using DDRT technique and immaturity seed of soyben,analyzed differential genes expression pattern between 20 hybrids and theirs parents in diallel cross of soybean.Analyzed relation eleven agronomic,two quality traits representation of hybrid and heterosis,the result showed that have obvious expression of genes differentially between hybrids and their parents.The correlation analysis showed that differential genes expression pattern and representation of hybrids,heterosis have some relativities.For six differential gene expression patterns and the eleven agronomic traits carry on the correlation analysis,The results showed that there were extremely negative correlation between plant height,No.of nodes per stem and the 110(The correlation coefficient respectively is -0.73**,-0.81**),there are extremely positive between protein and 110(The correlation coefficient is 0.58**);have extremely positive between plant height and 011(The correlation coefficient is 0.62**),No.of nodes per stem,100-seed weight,yield per plant and 011 existed negative rlation(The correlation coefficient respectively is -0.44**,-0.45**,-0.47**);nodes per stem and 101 have extremely positive rlation(The correlation coefficient is 0.71**),protein and 101 existed extremely negative rlation(The correlation coefficient respectively is-0.62**);stem diameter and 100 have negative rlation(The correlation coefficient respectively is-0.55*),fat and 100 existed positive rlation(The correlation coefficient is 0.43*).For six differential gene expression patterns and the MPH of eleven agronomic traits carry on the correlation analysis,there are four achieve at remarkable level in correlation coefficient.Among them,the BPH of plant height have positive rlation with 011(The correlation coefficient respectively is0.51**);the BPH of No.of branches per plant,No.of seeds per plant,yield per plant and 001 existed positive rlation(The correlation coefficient respectively is 0.46*,0.43*,0.47*).For six differential gene expression patterns and the BPH of eleven agronomic traits carry on the correlation analysis,there are four achieve at remarkable level or extremely remarkable level in correlation coefficient.Plant height,No.of nodes per stem,protein and 110 existed extremely negative rlation(The correlation coefficient respectively is-0.75**,-0.68**,-0.75**);plant height,rate of insect food,protein and 011 have extremely positive rlation(The correlation coefficient respectively is 0.59**,0.60**,0.59**);plant height,NO.of nodes per stem,protein and 101 have positive rlation(The correlation coefficient respectively is 0.52*,0.54*,0.52*); stem diameter and 100 existed negative rlation(The correlation coefficient is -0.47**),100-seed weight and 100 have positive rlation(The correlation coefficient respectively is 0.47*).Eighteen differential expressed cDNAs were cloned.By comparing to the homologous genes in GenBank,it was found that eight kind of enzynes,two is ligase,two is dehydrogenase,one is dioxygenase,one is endonuclease,one is helicase;yeast elongation factor complex Eefla,accommodate protein synthesize and promote apoptosis function;P97VCP,take part in protein degradation and regulate cycle of cell;activation of plant immunity by bacterial effector protein avrpto,take part in defense mechanism of plant;TIP30/CC3 gene,presume maybe have proactive function for diseaseresistance of plant;one of gene unkown.
