McroRNAs(miRNAs) are an class of single-strand noncoding RNAs with about 21-nucleotide length.Many of these are endogenous and evolutionarily conserved.MicroRNA play important roles in post-transcriptional gene regulation.Plant miRNA control many physiological processes by either cleavaging target mRNA or inhibiting its transtation.MicroRNAs are involved in plant development,signal transduction,protein degradtion,response to environmental stress and pathogen invasion,and regulate their own biogenesis.Research of plant miRNA is very fast.There are more than two hundreds of kinds of these have been discovered from the first identification in plant in 2002.Plants have many complicated mechanisms to adapt to all kinds of environment.In molecule level,they regulate many related gene expression to counteract enviroment sress in transcription and post-tanscription.Abiotic stresses such as mechanical stress,dehydration,salt,cold stress and so on modulate some special genes expression in transcription or post-transcripin and the genes improve the plant resistance.There are many problems to solve in our soybean production which has affected the yield and quality of it.This research identificate novel miRNAs related to resistance and their target and regulation pattern.Analysis on the influence that miRNAs to the soybean resistance will offer theory and technology to imrove soybean inheretance and novel genes discovery and set a start of the soybean miRNA.The research get sequences of the soybean miRNAs which was predicted by bioinformatios by searching the soybean miRNA database.11 miRNAs were cloned and obtained theier sequences.The research predicted the miRNAs' targets seaching by the keys of miRNA mature sequence based on the complementarity between miRNA and mRNA and identified the cleavage of the target of miRNA157 for 10 times.The mir160 promoter was cloned and we found a ARF(auxin response factor) by analysis on its cis-acting regulatory DNA elements.That indicates miRNA160 is realtaed to stress.Finally,i analyse the miRNAs' expression by norhtern blot.
