Morphological And Distributive Studies Of Haemocytes Of Patinopecten Yessoensis

The morphological characterization and functions of haemocytes of Patinopecten yessoensis are studied by histological and histochemical methods in this paper.Light(vivisectional observation,Glutaraldehyde disposal,Giemsa and HE staining method) and transmission electron microscopical studies are carried out in order to characterize the haemocytes of the bivalve mollusc,Patinopecten yessoensis.According to nucleus and cytoplasm characters,three types of haemocytes are observed by light microscopy.The typeâ… haemocytes are small,round,show high nucleus ratio.The nuclei are round or oval.The cytoplasms are little and they are in the verge of the nucleus.Rich organelles are found in the typeâ…¡haemocytes cytoplasm.This type haemocytes is amoeboicyte,shows low nucleus ratio.The quantity and nucleus ratio of the typeâ…¢haemocytes are between the typeâ… and the typeâ…¡haemocytes.According to the vivisectional observation,the typeâ… haemocytes have no pseudopod;the typeâ…¡haemocytes have a lot of lamellipodial protrusions;the typeâ…¢haernocytes have some filopodia-like projections.According to the observation of Giemsa staining method,the typeâ… haemocytes are basophil; The pigmentation of the typeâ…¡haemocytes is light-colored;the pigmentation of the typeâ…¢haemocytes is darkest.The shades of different budding typeâ…¢haemocytes are different.According to the observation of transmission electron microscopy,the typeâ… haemocytes are round or oval.The nuclei are large.The cytoplasms are little.The typeâ…¡haemocytes are variform.They have erose edges and protuberances.The nuclei are round,oval or kidney-shaped.The cytoplasms of the typeâ…¡haemocytes are abundant,containing many mitochondfion,endoplasmic reticulum segments,vesicle and glycogen.The typeâ…¢haemocytes are round or oval,have protuberances,the cytoplasms are rich,containing some mitochondrion,endoplasmic reticulum segments, vesicle and glycogen;some haemocytes are effete,organelles dissolve,nucleus shrink slightly.According to the results,these haemocytes are lack agranular haemocytes.According to the observation of light and transmission electron microscopy,there is no distinct granule in the cytoplast of Patinopecten yessoensis haemocytes.According to the HE staining,the haemocytes mostly consist in mantle,gill, alimentary canal and adductor muscle of Patinopecten yessoensis.In these tissues,the typeâ…¡haemocytes are most.They are general distributing in the gaps of epidermis and the connective tissue near the epidermis.The typeâ…¡haemocytes have a high phagocytosis.In the mantle,the haemocytes mostly distribute in the connective tissue. A few of haemocytes dissociate at the gaps of epidermis cells.There are a lot of hemocoels and abundant haemocytes in the middle zone of mantle.The haemocytes are dense and diversiform.Many intergradational haemocytes are observed.Many haemocytes are in the hemocoels and the connective tissue of gill.The haemocytes in the hemocoels of main gill are dense and abundant.They largely infiltrate to the connective tissue of gill.The haemocytes of common gill are less,many of them are infiltrating to the top of the epidermis.
