In this study,a molecular testing procedure was established based on SSR and AFLP marker,maize germplasm databases were constructed and the feasibility of the application of the molecular testing procedure in the identification,evaluation and protection of maize germplasm was proved.The main results were as follows:1.This research optimized SSR and AFLP marker in DNA extraction,PCR amplification,gel electrophoresis,etc.and established a systemic and local molecular testing procedure.2.Based on polymorphic and stable bands,15 primers were screened out of 20 primers.DNA fingerprinting databases of 92 common maize inbred lines in China and 160 important maize hybrids in Northeast of China were constructed by 15 primers.15 primers produced 82 alleles with the average of 5.47 per loci and a range from 3 to 9.The PIC per loci had a range from 0.5836 to 0.8607 with the average of 0.7229.3.Based on polymorphic and stable bands,7 combinations of MseI and EcoRI primer were screened out of 64 primer combinations.DNA fingerprinting databases of 22 representative maize hybrids were constructed by 7 primer combinations,which were cultivated by Jilin Academy of Agriculture Science.The PIC per primer combination was more than 0.98.Shannon-weaver index per primer combination had a range from 4.1647 to 4.7326.4.Based on the established technical system,the results showed that SSR markers could be completely used to the identification of maize germplasm could be used to the disease-resistant evaluation of maize germplasm combined with field trails and AFLP markers converted into SCAR markers could be used to protection of maize germplasm.
