Chinese fir(Cunninghamia lanceolata (Lamb.)Hook) is one of the native evergreen coniferous species to China. And it have had been widely cultivated through all the Southern China, for its perfect characteristics such as fast growing, high quality of wood properties. Currently somatic embryogenesis (SE) system has been an important means of large-scale production in Chinese fir.We presented here the results, with developing seed of Chinese fir at different stages, to reveal the characteristics of proteomics and metal elements during seed developmental processing. The 2D difference gel electrophoresis was preformed to identify the changes in the proteomic profile upon the developmental stages. There were 153 interest spots to be found through the analysis. Some proteins, identified by LC/MS/MS, were expressed increasing in the cotyledon phase. The legumin-like storage protein, Lea protein, PR10-1.13, small heat-shock protein seems, was to be thought to relate to the zygotic embryo development processing, respectively. The key elements in the developing zygotic embryo and female gametophyte of Chinese fir were analyzed with inductively coupled plasma emission spectroscopy also. And during the seed development, the element Cu was increased gradually in small amount for all the genotypes tested. But P was increased in larger amount; K and Mn was going down first and up later; the patterns for Ca and Mg seems to be a little bit complicated, however, One strong proof was shown that Ca was reached to the highest peak in the early stage of seed development, and then turn down to the lowest, whereas, the Mg reached to the highest peak at later stage of embryo full. The Co and Ni were minor among all the stages.
