Excessive intake of fluorine may bring about fluorosis, cause multiple organs, multifunctional skeletal and nonskeletal lesion. In nonskeletal lesion, immune toxicity of fluorine has always been more attention.by people. Bovine spleen lymphocytes were used as subjects in this study in vitro, and different concentrations of NaF were added into culture medium, then membrane permeability, membrane protein's amount and components, sialic acid and cholesterol content, antioxidant function, cytokine content, IL-2RmRNA and IL-6RmRNA were measured, in order to investigate the effects of fluorine on lymphocytes membrane structure and function. The results showed that:1. Protein component of lymphocytes membrane were analyzed, and the result demonstrated that protein bandsⅠ,Ⅱ,Ⅲ,Ⅳ,Ⅴpercentage content increased and protein bandsⅥ,Ⅶ,Ⅷpercentage content decreased, the results showed that membrane protein crosslinking caused by fluorine, and membrane macromolecular structure was destructed, then resulted in protein component changed.2. Leakage rate of lymphocytes lactate dehydrogenase (LDH) increased, then resulted in cell viability decreased, the results showed that lymphocytes membrane permeability could be changed by fluorine.3. Lymphocytes membrane sialic acid content decreased, the results showed that lymphocytes membrane structure could be destructed by fluorine.4. Lymphocytes membrane antioxidant function was measured, the results showed that membrane glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) activity decreased, membrane lipids peroxide malondialdehyde(MDA)increased, the results demonstrated that lymphocytes membrane could be resulted in oxidative damage, lead to oxidative stress, and then played to immune membrane toxic effect.5. Na+-k+-ATPase, Ca2+-ATPase and Mg2+-ATPase activity of lymphocytes membrane were decreased at different degree, the results showed that ATPase activity of lymphocytes membrane was inhibited by fluorine.6. Cholesterol content of lymphocytes membrane was increased, P value of membrane fluorescence polarization degree andηvalue of viscosity increased, the results showed that lymphocytes membrane fluidity could be decreased by fluorine.7. The secretion of IL-2 and IL-6 of lymphocytes were decreased by fluorine, then IL-2RmRNA and IL-6RmRNA expression were inhibited. The results showed that cytokine content secretion and receptors expression were inhibited by fluorine and then immunologic cytotoxicity was exerted.Immunologic cytotoxicity of fluorine was investigated from subcellular and molecular levels, and it can offer the theoretical and experimental basis for clarification of immune cells lesion and immunosuppressive mechanism.
