Effects Of Chinese Herbal Medicinal Monomer,Zinc,Selenium On Proliferation And Secretion Function Of Swine Spleen Lymphocytes In Vitro

Study the effects of Astragalus polysacharides(APS),Acanthopanax senticosus saponins B(ASS B)Chlorgenic Acid(CHA),Lobetyolin(LT),Ginsenoside F3(GS F3),ZnS04,Zinc glycinate(ZnGly),Na2Se03 and Selenomethionine(SeMet)on proliferation and secretion function of swine spleen lymphocytes in vitro.CCK-8 assay for the proliferation of lymphocytes,ELISA methods were used to detected IL-2.IL-4 and IL-10 levels.The results showed that:①Astragalus polysaccharide can promote swine spleen lymphocytes proliferation independently or together with ConA or LPS(P<0.05),the best level of APS is 100 or 200μig·mL-1;Acanthopanax senticosus saponins B can promote swine spleen lymphocytes proliferation independently or together with LPS(P<0.05),the best level of ASS B is 50 μg·mL-1;Chlorogenic acid and Lobetyolin can promote swine spleen lymphocytes proliferation independently or together with ConA or LPS(P<0.05),the best level of CHA or LT is 200 μg-mL-1;Ginsenosides F3 can promote swine spleen lymphocytes proliferation together with LPS at the concentration of 5 and 20 gg·mL-1(P<0.05).②Astragalus polysaccharide can promote swine spleen lymphocytes secrete IL-4 and IL-10 at the concentration of 50μg·mL-1;Acanthopanax senticosus saponins B can promote swine spleen lymphocytes secrete IL-2,IL-4 and IL-10 at the concentration of 50 μg·mL-1;Chlorogenic acid and Lobetyolin can promote swine spleen lymphocytes secrete IL-2,IL-4 and IL-10 at the concentration of 200 μg-mL-1;APS+ CHA and ASS + CHA can promote lymphocyte secrete IL-2,IL-4 and IL-10.③ZnSO4 can not promote swine spleen lymphocytes proliferation significantly(P>0.05),Zinc glycinate can promote swine spleen lymphocytes proliferation significantly at the concentration of 40 μm·L-1(P<0.05);Na2SeO3 can not promote swine spleen lymphocytes proliferation significantly(P>0.05),Selenomethionine can promote swine spleen lymphocytes proliferation at the concentration of 0.05,0.1 and 0.5 μmol·L-1;ZnGly&SeMet combination can promote swine spleen lymphocytes proliferation in vitro.④Zinc glycinate can promote swine spleen lymphocytes secrete IL-2 at the concentration of 20 μmol L"1(P<0.05),Zinc glycinate can promote swine spleen lymphocytes secrete IL-4 at the concentration of 40 μgmol·L-1,(P<0.05),Zinc glycinate can promote swine spleen lymphocytes secrete IL-10 at the concentration of 20 and 40μmol-L-1(P<0.05);Selenomethionine can promote swine spleen lymphocytes secrete IL-4 and IL-10 at the concentration of 0.1 and 0.5 μmol-L-1(P<0.05),can not promote swine spleen lymphocytes secrete IL-4 significantly(P<0.05);ZnGly&SeMet combination can promote swine spleen lymphocytes secrete IL-2,IL-4 and IL-10 in vitro(P<0.05).In summary,Astragalus polysaccharide,Acanthopanax saponins B,Chlorogenic acid,Lobetyolin,Zinc glycinate and Selenomethionine could promote swine spleen lymphocytes proliferation in vitro,and could regulate of the immune system by promoting lymphocytes secrete IL-2,IL-4 and IL-10.
