| The cucumber, for instant consumption, rich nutrients and people love it.Different size and shape cucumber meets the need for kind of instant consumption, cooking and processing.In this study, use of phytotomy and flow cytometry, by means of four varieties of cucumber types (South China type, North China type, the European greenhouse type, pickled type) and monolinic cucumber to split and expansion of the process,cell morphology, cell cycle and cell proliferation, a comprehensive comparative analysis of cytology from the perspective of cucumber fruit-depth understanding of the dynamic changes of growth and development, in order to carry out the cytology of cucumber fruit development and lay the foundation for molecular biology research.1. The study set up growth curve models of different types of cucumber. The growth curve consistented with'Y=a+bX+cX2+dX3'.The first inflection point was fast period of cucumber fruit'RGR, and also was key period of cucumber's production and quality which were building up. D0422 and D0401's maximum RGR were in the third days to the fifth days. 649 and Jinchun5's maximum RGR were in the fifth days to the seventh days. D0118-2yuan's maximum RGR were in the ninth days to the eleventh days. D0118-2chang's maximum RGR were in the third days to the fifth days.2. The cell division is a persistent process. The pericarp and sarcocarp's S and G2/M phase had the same cell cycle in flowering day. Both of them had a coincident saliens punctum and the same vigorous cell division. After this time, there cell cycle was different. There was 1~2 S and 2~3 G2/M crest-time. The crest-time value of pericarp's S and G2/M consistented with the fruit shape and RGR's. Changs of spatial cell division disacorded, but changed samely the temporal. Fruit's length speed faster than diameter before the eleventh day, and diameter increased accompanied by length. The cell shape changed unclearly. It was obvious that cell splited along fruit length direction. At the same time, cell division gone with cell enlargement. Cell enlargement was in the ascendant when cell division lasted out a period of time. The increasing speed of f ruit diameter faster then length.3. The sarcocarp enlargement extent was greater than pericarp's at cell ray and tangent direction. Cell enlargement extent and crest-time were coincident about a great number of cucumber sarcocarp, and therefore cell shape was rotundity or oval. Cell enlargement extent and crest-time weren't coincident about a great number of cucumber pericarp, and therefore cell shape was rectangle. A great number of pericarp cell has two obvious enlargement period. Cell enlargement had relations with fruit length. The ray direction cell diameter and fruit diameter, tangent direction cell diameter and fruit length represented positive correlation. The correlation of fruit cell diameter and length and diameter exceeded pericarp cell diameter and fruit length and diameter.Furthermore, the growth prophase of cell diameter and fruit length and diameter larger than anaphase; however, the range of reducing on anaphase of sarcocarp cell diameter less than pericarp's.4. Changes of the four endogenous hormone content was: the seventh days fruit's ZR and GA3 greater than the fifteenth days, the fifteenth days fruit's IAA and ABA greater than the seventh days.The endogenous hormone content of pericarp greater than sarcocarp or a little.The content change was GA3> ZR > IAA >ABA. Cell division bloomed at fruit development prophase, and declined at anaphase.The ZR content had the same orderliness.The IAA content of all variety cucumber retained a high level, and made cell enter G1.The GA3 content of all variety cucumber had the same transformation.The time of pericarp cell division was longer than pericarp. At this time, the GA3 content of pericarp was higher than sarcocarp.The law of ABA content was not clearly at pericarp and sarcocarp.All the variety had a great contrast. Most varieties of fruit for two days, for the pericarp cell splited stronger than sarcocarp. It reacted on the growth of the fruits of "brake" balanced regulation. |
