For seeking new friendly nematicide for controlling Meloidogyne incognita Chitwood, nematicidal constituents in water extract of MeOH-Ext. from Chryanthemum coronarium were studied with bioassay in present paper. The results would provide some theoretical basis for the research and development of friendly nematicides. The results could be summarized as follows:1. The differences among the extractive rate and nematicidal activities of the extracts with cold dip and blitzkrieg extraction were not significant. Blitzkrieg extraction could substitute cold dip for extacting the nematicidal components from Chryanthemum coronarium.2. The differences among the extractive rate and nematicidal activities of the extracts with water and methanol were significant. Although the nematicidal components in Chryanthemum coronarium existed in its water extract of MeOH-Ext., it is not suitable to use water replacing methanol as the extraction solvent.3. In vitro bioassay results showed that, with 1% of the proportion of five extracts of MeOH-Ext. from Chryanthemum coronarium to treat the eggs of Meloidogyne incognita Chitwood, the sequence of the restraining effects of those extracts to the embryonic developments of Meloidogyne incognita Chitwood from strong to weak showed as follows: water extraxct, n-BT- Ext., TrOEt-Ext., EtOAc-Ext., PtOEt-Ext..The restraining effects showed in the developing process of treated eggs slower than the comparison. With 1% of the proportion of water extract to treat for 264h against Meloidogyne incognita Chitwood, only 15% eggs had completed their embryonic developments.4. In vivo bioassay results showed that, with 1% of the proportion of five extracts of MeOH-Ext. from Chryanthemum coronarium to treat the J2, the sequence of the restraining effects of those extracts to the post-embryonic development of Meloidogyne incognita Chitwood from strong to weak showed as follows: water extraxct, n-BT-Ext., TrOEt-Ext., EtOAc-Ext. and PtOEt-Ext.. The restraining effects showed in postponing the process of the post-embryonic development of Meloidogyne incognita Chitwood. The restraining effects mainly influenced the developing processes of J2 and female, while they had no significant influence on the development stages of juvenile 3 and juvenile 4.In addition, the influences of different extracts agaist the post-embryonic development of Meloidogyne incognita Chitwood also showed in postponing juvenile penetration and female reproduction, disrupting or inhibiting the amount of eggs by female. The water extract was the most effective one among the tested five extracts to the tested nematode. The post-embryonic development, juvenile penetration, female reproduction were delayed 4, 5, 9 days respectively. Meanwhile, the extract disrupted or inhibited the amount of eggs by female significantly.5. Based on the results of bio-screening, the nematicidal ingredients in water extract of MeOH-Ext. from Chryanthemum coronarium were investigated thoroughly. The extract was purified by Silica gel column chromatography and pre-LC, two compounds were obtained finally.By combining the related reference with the data of NMR and IR, the structures of the two compounds were elucidated. H1 was Parthenolide; H2 was Hyperoside.Both of H1 and H2 were first isolated from Chryanthemum coronarium.
