| Objective: To investigate the leftover in the processing of animal productions, in this research, the Yak bone of Gannan was used as raw material. At first, the bone was refin- ing after have been cooked at high temperature and pressure, then the best enzymatic hy- drolytic processing parameters of bone collagen was determined by indexed with the content of peptides and the hydrolysis rate of protein through single factor and orthogo- nal test. Then the rats were fed of nutrition bone powder that was made the spray drying ultrafine bone powder with the casein phosphopeptide (CPP) and inulin. The first test group bone powder 75%,CPP5%, inulin 20%; the second test group bone powder 75%, CPP10%, inulin 15%, and the control group at the same level of calcium. After the feed- ing test, the bone calcium content was analyzed through the atomic spectrophotometer; the bone density was determined by the Dual Energy X-ray Absorptiometry; the serum biochemical indexes (Alkaline Phosphatase, AKP; triglyceride, TG; cholesterol, CHOL and K+,Na+,Cl-,Ca2+)were measured by electrolytic analyzer. The results showed as following:1,The content of peptides and hydrolysis rate of protein was 5.358mg/mL, 5.108mg /mL,4.783mg/mL and66.98%,63.85%,59.79%respectively, when the bone collagen was hydrolyzed by Papain, Bromelain and Ficin. The results showed that: the hydrolysis abi- lity of Papain was stronger than the others, so the papain was used in the test.2,The range of factors were determined through single factor test. The enzyme dosage 2500~3500U/g,enzymolysis time 6~10h,temperature 50~60℃, pH 5~7.3,The best processing parameters were obtained through orthogonal test of L9(34). The parameters as following: temperature 55℃,pH5.0,enzyme dosage 3500U/g,enzymol- ysis time 6h.The content of peptides was 5.845mg/mL and the hydrolysis rate was 73.06%that was determined through the biuret method.4,The test of feeding showed that: the two test groups of rats were developed norm- ally. The average food intake per day of two test groups was 13.57g/rat,14.21g/rat; the weight increased 150.6g/rat,142.83g/rat; the femur weight, food intake per day and weig- ht increased of the test groups all significantly greater than control group(P<0.01). 5,The bone density of two test groups were 23.48%,20.0% higher than that of the con-trol group, and the difference was extremely significant (P<0.01).The content of bone calcium of test was 8.56%,5.79% higher than that of the control. The results sho- wed that: the effect of supplemented calcium was obvious in test groups6,The tests groups average content of AKP and TG in serum compared with the control there was no statistical difference(P>0.05); and the average content of CHO was 2.090mmol/L, 1.855mmol/L, the second test group was 9.82% lower than that of the control (P<0.05).7,The average content of K+,Na+ there was no differrence among groups. But the content of Ca2+ in serum was 2.873mmol/L,2.653mmol/L, and 4.71%, 12% lower than that of the control, the difference was extremely significant (P<0.01, P<0.05).The content of Cl- in serum of test group was 116.641mmol/L,111.883mmol/L, the difference was significant in the second group(P<0.05).8,The results of the test showed that: the rats that intake the nutrition bone powder, it's bone calcium and bone density both increased, and there was no effect on the AKP, TG, CHO and the balance of electrolytes in serum, so the nutrition bone powder was a kind of natural supplement of calcium. |
