Screening And Biological Characteristics Of Metarhizium Anisopliae Strain Highly Pathogenic To Haemaphysalis Qinghaiensis Ticks

Metarhizium spp are important group of entomopathogenes, and can be used to control lots of pests in agriculture and forest. The biocontrol method which using Metarhizium as a biocontrol agent is an important measure to control ticks. In order to get some high virulent strains which are fit for using in the field, and explore the efficient ways of using Metarhizium to control the pests, the listed researches below were done:Metarhizium strains were isolated from dead body of Haemaphysalis longicornis tick, and then identified and named MaLS01. In order to screening for some virulent and excellent strains, the pathogenicity against Haemaphysalis qinghaiensis were tested. In order to determine the best cultural conditions, the cultural characteristics of Metarhizium MaLS01 were tested. The capability of Metarhizium MaLS01 with five pesticides were tested to evaluate the covalent feature, and the solid-state cultural medium were also screened. The results were the below:1 The isolation and identification of Metarhizium anisopliaeA strain of Metarhizium anisopliae was isolated from dead body of Haemaphysalis longicornis tick, then identified by observing cultural characteristics on PDA medium, the ITS1-5.8S-ITS2 sequence was sequenced, and named MaLS012 The filter of strain highly virulent to Haemaphysalis qinghaiensisThe virulence of seven different strains of Metarhizium anisopliae and Beauveria bassiana to engorged females of Haemaphysalis qinghaiensis were tested, The results showed that the cumulative mortality rates of Metarhizium anisopliae and Beauveria bassiana strains were 80%,66.67%,60.00%,63.33%,55.67%,43.33%,33.33% when the spore concentration was 1×108/ mL. The Metarhizium anisopliae strain MaLS01 showed high virulence to Haemaphysalis qinghaiensis.3 The cultural characteristics of Metarhizium anisopliae strain MaLS01The Metarhizium anisopliae strain MaLS01 was cultured on five different medium, and the suitable temperature,pH,C/N were determined. The results showed that this strain grew well on PPDA culture medium, the most suitable culture condition is temperature 30℃,PH 5～9 and C/N 2:0.5～1.4 The capability of Metarhizium anisopliae strain MaLS01 with pesticidesThe capability of Metarhizium anisopliae strain MaLS01 with five pesticides were tested, The results showed that this strain had well capability with Abamectin. The spore germination rates were 85.29%,90.00%,96.44% for 12 hour.5 The solid-state fermentation of Metarhizium anisopliae strain MaLS01Selected rice,wheat bran,cavings,lucerne and indian meal as feed, designed A,B,C,D,E,F six medium to filter best solid-state medium, the results showed that strain grew well on medium E, that could be used as solid-state fermentation for Metarhizium anisopliae strain MaLS01.
