Study On The Pharmacology And Toxicology Of Pyroline And The New Preparations Of Pyroline

The new drug pyroline which is the effective constituent in pyrolae and its new preparations were developed successfully in Lanzhou Institute of Animal Science and Veterinary Pharmaceutics Science of Chinese Academy of Agricaltural Sciences. There are only few data reported about pyroline and the new preparations recently. The study was carried out to comprehensively evaluate pharmacology and toxicology of pyroline for its further clinical application.By determing the minimal inhibitory concentration and bacteriostasis diameter, the results showed that the bacteriostasis effect of pyroline was very good. Streptococcus mastitidis, Staphylococcus aureus and Bacillus aeruginosus were responsive to pyroline than Bacillus coli, Streptococcus dysgalactiae and Bacillus proteus .Bacteriostasis effect of pyroline and ciprofloxacin to Bacillus coli and Bacillus aeruginosus in vivo was determined by using infected animal model under the experimental results above. The group treated with pyroline was cured in total, the group treated with ciprofloxacin (Bacillus aeruginosus infected animal model) was cured by 95%.The results indicate that bacteriostasis of pyroline in vivo was very strong and better than ciprofloxacin in animals infected by Bacillus.The acute and subchronic toxicity experiments were performed to evaluate safety of pyroline and its new preparations. The results showed that the LD50 of pyroline was 513.25±0.023 mg/kg,the LD50 of the new preparations of pyroline was 666.45±0.0222 mg/kg which indicates that pyroline and its new preparations were low poisonous drugs.5. Subacute toxicity experiment was proceeded to evaluate the security of pyroline and the its new preparations. The results showed that the dead phenomenon wasn′t appear,and that there no abnormal phenomenons in the three giving medicine groups,including clinical representations,body weight increasing,physiological index of blood,biochemical indicator of blood and index of organs.There were no abnormal phenomenons in the low and medium dose groups of histological structures of organs.There was slight pathological changes in the high dose group of some rats of histological structures,including Liver and Kidney.The method and the results of subacute toxicity experiment of the new preparations of pyroline were the same as of pyroline. It can be drawn conclusion that pyroline and its new preparations were low poisonous drugs.Above the results of pharmacodynamics,acute and subacute toxicity experiments confirmed that pyroline and the new preparations of pyroline were a kind of medicine which had broad spectrum,high performance and low negtive effects on animals.It should be widely apply in the clinic.
