Superoxide dismutase is an important enzyme of the metal which exists in various tissues of Organisms.The main role of Superoxide dismutase is removal the superoxide anion (O2-) specifically and balance oxygen free radicals in Organisms . So as to avoid the serious damage to protein, lipid, DNA and other cellular components by the accumulation of reactive oxygen species. In accordance with its combination of different metal ions Superoxide dismutase can be divided into three kinds, namely: Cn / ZnSOD, Mn SOD and Fe SOD. And Cn / ZnSOD is divided into two types :One is the intracellular called copper-zinc superoxide dismutase (Cn / ZnSOD); the other is known as the extracellular called extracellular copper-zinc superoxide dismutase (EC-SOD ), they have the same disproportionation reaction rate and the similar kinetic constants. As a biological detergent Of free radicals in organisms ,the studying of superoxide dismutase (SOD) has important significance in theoretical meaning and practical application. In the theoretical sense,the research of superoxide dismutase play a important role in understanding the homology between species and evolution of the relationship more clearly. In the sense of practical application,the research of superoxide dismutase,especially the function and activity research of SOD protein and it's mimics will greatly contribute to the application of superoxide dismutase in medicine and health care, food industry, cosmetics, agriculture and other the fields,in order to serve better for our common humanity.In recent years, there are several biological SOD genes have been successfully cloned , expressed and functional analysised ,in the study of Superoxide dismutase in insects of silk Yamamoto K (2005) and Cong Haifeng (2007)reported the sequence and related functions of Bombyx Mori and Bombyx mandarina respectively.So far, the SOD gene of Antheraea pernyi has not been resolved yet , therefore, this experiment cloned Antheraea pernyi copper/zinc superoxide dismutase (Cu / ZnSOD) gene sequence of the ORF sequence by RT-PCR method firstly .This sequence encodes 154 amino acids with Cu / ZnSOD conservative structural characteristics. The evolutionary analysis shows that homology identities between Antheraea pernyi,Bombyx mori,Hyphantria Cunea,Bombyx mandarina,Drosophila nasuta are 81.5%,81.7%,81.5%,66.7%, respectively. In addition, the expression of Cu/Zn SOD increases after 5 min of UV radiation, reaching to the peak at 10 min and decreasing at 15 min after treatment..The research of this experiment provides strong conditions in researching the structure ,function, gene expression and regulation mechanism ,as well as phylogenetic of Cu / ZnSOD gene in Antheraea pernyi model .
