This study was conducted to explore the effects of dietary of different protein type and protein level on spring multiplication of Apis cerana cerana. and revealed the expression profile of copper/zinc superoxide dismutase gene of Apis cerana cerana (Acc-SOD1) by molecular biology methods.The effect of dietary of protein type of three kinds on spring multiplication of Apis cerana cerana were studied. The result showed that there is no significant difference among the experimental groups and control group (Group A) in the amount of sealed brood and honeybee population (P>0.05). The emergent weight of workers in group B, which was fed with the mixture (pollen, cooked soybean, yeast extract, sugar) was better than that of the Group C and D (P<0.05), while it is similar to the control group (P>0.05).We studied on the effect of dietary of different protein levels on spring multiplication and larva oxidative stability of Apis cerana cerana. The results showed that the effect on the amount of sealed brood and population with feed containing25%-30%protein was smaller than that of others.there was no significant effect on the emergent weight, protein content and T-SOD activity of larva among the feed containing20%-35%protein and the control group (P>0.05), yet the T-SOD activity of control group were significant higher than that of the test group of15%protein level (P<0.05). With increasing protein level, there was no significantly effect on the MDA content of larva (P>0.05).Analysised this genetic structure and function based on cloned Acc-SOD1gene, the expression level of Acc-SOD1in different tissue and at different developmental stages (3rd and6th instars larva,1st and4th pupa and1st and7th adult), as well as in50mg/kg and500mg/kg vitamin C treated bees, were detected by qRT-PCR, and calculated the mortality rate of bees after vitamin C treated. The full length of Acc-SODl cDNA (GenBank accession number is JN700517) was631bp, encoding152amino acids, predicted MW and PI were15.65kD and6.21. The deduced amino acid sequence of Acc-SOD1showed68%-99%identity with that of other insects, respectively. The expression level of Acc-SOD1was significantly changed at different developmental stages, which peaked in6th instars larva, then started to drop dramatically in4th pupa, and almost no change can be detected after being adult, the expression level of Acc-SOD1in the brain and abdomen was significant higher than that in thorax (P<0.05). Moreover, with the increase of the vitamin C concentration, the mortality rate of bees decreased significantly (P<0.05), but this expression level of Acc-SOD1was no significantly changed (P>0.05)...
