Potato (Solanum tuberosum L.) is the forth in agriculture and industry in both China and the world. Late blight caused by Phytophthora infestans (Mont.) de Bary influence the production of potato all along. It becomes urgent to improve late blight resistance level by means of breeding methods long last resistance potato varities. Horizontal resistance is controlled by many interacting genes. Many late blight resistances QTL have been mapped on all of the 12 chromosomes of potato. However, it remains unknown which genes are the really contribute to horizontal resistance, which brings some difficulties for breeding via molecular marker-assisted selection (MAS).In present research, based on the former result, we use the candidate gene(CG) strategy to map our CG on the linkage of potato and comparie their location with known QTL, That will help for to find the real genes control horizontal resistance to late blight, constitute basis to MAS. The main results are as following:1. Eighty-six pair primers were designed according to 38 CGs. Of which, 19 pair primers from 18 CG amplified 27 polymorphic bands in BCT and PCC1 population from(BCT has 22 bands and PCC1 has 5). 12 bands were from our 11 CG. Other 15 bands were amplified from other sequence of the popato genome, in which 9 bands can find homologous sequence from NCBI and 6 can not.2. The data of newly find polymorphic bands was integrated into original data of the BCT and PCC1 population and maped again. 24 in 27 bands were mapped on the chromosome of the two populations. 11 genes were mapped on BCT female chromosomes ofâ… ,â…¢,â…£,â…¤,â…¦,â…§andâ…¨, 11 genes were mapped on BCT male chromosomesâ… ,â…¤,â…¥,â…©,â…ªandâ…«and the other two were mapped on PCC1â…§andâ…«chromosomes.3. The comparison of mapping result and the known QTL indicate that 07-F08-P1-564 located in the late blight resistance QTL; 11-A04 and TC128268 were land in tuber dormancy and secondary metabolism related QTL; NAC was mapped to a QTL region relatived to tuberization and insect resistance; 10-B01 located in a QTL of insect resistance; Both PK4-P1-425 and 07-F08 were put into a QTL region relatived to tuber dormancy.Although all the primers were designed from late blight resistance related CGs, one gene located in late blight resistance QTL, others located in the QTL region rather than late blight resistance QTL. The main reason may be due to so many primers show no polymorphism in two populations we used. The new makers found in present studies will provide foundation for further developing molecular makers and insight researchs on related potato agronomy traits.
