| Estrogen play an important role in the development of the early fetal mammalian gonads, also at the same time, a majority of mammalian gonadal secret androgen,and androgen was converted into estrogen under the effects of aromatase which were widespreadly distributed in the gonads, thus affecting the development of the gonads Pig has been a model which is widely used as in human beings medical research and a important livestock animal, thus the study is very important to understanding the changes of the early fetal mammalian gonadal through the fetal pig gonadal changes.The present study was undertaken to investigate the tissue structure chang of porcing feat ovary/testis(gonad) at 33rd,40th,47th,54th,61st day of gestation by hematoxylin and eosin(HE) staining. And based previous experiments, the localization of ERαand ERβwere observed by immunohistochemistry, and then the results were analysed and summarized. The experimental results are as follows:HE staining:①Porcine gonads of 33rd day of gestation had been differentiated, and in the male gonad, the albuginea and tesis cords were observed, in contrast, the medulla cords were observed in the female gonad.②The testis cords was much more slender and shorter at 40th day of gestation and some has formed arc-shaped, when synplasma-like cell groups occurred in the ovary and the cord-like structure disrupted in the medulla.③the testis cord has further degenerated, the majority have the form of arc-shaped lumen, medullary cords were broken into fragments to be island-like cell groups or primordial follicle.④At 54th day of gestation, seminiferous tubule has formed, the lumen has no gap, when in the ovary, the cortex and medulla could be clearly distinguished.⑤At 61st day of gestation, the testicular seminiferous tubules has been further developing, the lumen gap expanded markedly, and some proimordial follicle were ovserved and primordial follicles occurred in deep cortex.Immunohistochemical result:①ERαwere immunoreactive at different period in the ovary. At 33rd day, ERαwas expressed on the surface epithelium and stromal cells in the oocyte nest, at 40th day, the immunoreactivity on the surface epithelium is very weak, and is much stronger on some oocytes in the nest, at 47th day,ERαwas observed in the surface epithelium, and on some of oocyte and the surrounding stromal cells in the oocyte nests, at 54th day, ERαwas expressed on the surface epithelium, oocytes in the oocyte nest and some fusiform-shaped stromal cells, to 61st day, ERαwas observed on the surface epithelium and oocyte in the follicle, especially the primordial follicles in the cortical part.. ②ERαwere immunoreactive at different period in the testis, on 33th and 40th day, ERαwere localized in the tunica albuginea and germ cell, on the 47th day , ERαwas present in the tunica albuginea,germ cell and some interstitial cell, when on the 54th day, was mainly present in the albuginea,germ cell and some interstitial cell and peritublar cell,but at 61st day, ERαwas expressed weakly in the tunica albuginea and interstitial cell, but stongly in the germ cell.③ERβwere immunoreactive at different period in the ovary. At 33th day, ERβwas present in some oocyte, at 40th day, was localized in the oocyte and some granular cell, at 47th day, ERβwas mainly expressed in the oocyte in the oocyte nest, and especially in the cortical part. At 54th day, ERβwas present in the oocyte and granular cell, but at 61st day, was much strongly in the oocyte, and weakly in the granular cell.④ERβwere immunoreactive at different period in the testis. On 33 days, ERβwas localised in the nuclus of germ cells. On 40th, 47th, 54th and 61st, ERβwas mainly present in the germ cell, and the immunoreactivety is increased gradually, but weakly in the tunica albuginea.This study systematically investigated the histological change of fetal ovary and testis of 33th,40th,47th,54th and 61st.Also by dectecting the distribution of ERαand ERβ. |
