This study was to explore the special aquatic vegetables Vallisneria barbed effective means of vegetative propagation of tissue culture in the process of disinfection methods, media types, explant selection, light temperature treatment, the choice of auxin and cytokinin etc. A comparative study. Buds sharp focus on the best procedures for tissue culture medium, the establishment of Vallisneria gill tissue culture system.The results showed that:1. Rhizome Vallisneria 6BArbed as the lateral bud explants, the choice of disinfection drugs showed that 70% alcohol to disinfect with 30s, and then increase 0.1% Mercuric chiorideand tween 20 (3~5 drops) disinfection of 12min, the best disinfectant;2. Vallisneria 6BArbed tip of the bud of the early generation of tissue culture, MS medium than the effect of N6 medium and the survival rate of growth is better, so choose as the 6BAsic medium MS; adding agar of about 7.5g, solid medium;3. Different explant organs, under the same conditions on the rhizome, leaf, bud tip for induction, shoot tip callus formed structure, the color green bias; bud tip the shortest number of days to start the 5d, followed by leaf 7d, the slowest start rhizome 15d; bud tip the lowest survival rate of the highest pollution rates were 77.5% and 20% complete and easy to cultivate plants; leaves and stem segments of the successful induction of callus induction rate is a low but the realization of the plants from the callus regeneration; therefore the best option explants lateral bud of the bud tip of rhizome;4.
