Polygonatum odoratum (Mill.) Druce florum, which belongs to Polygonatum plant of Liliaceae, has exploitation and utilization value as a kind of traditional food and a Chinese herbal medicine plant. However, there are many problems during the production process of polygonatum with large amount of seeds using, easily breeding quality degradation, and serious genetic separation. Therefore, investigations on tissue culture and rapid propagation in Polygonatum odoratum Druce have important theoretical and practical value for genetic improvement, standardized production and industrialization. In the paper, the effects of major factors on tissue culture and rapid propagation are studied to establish tissue culture systems in Polygonatum, which can provide very important reference for Polygonatum industrialized production, germplasm preservation and genetic improvement. The major results are as follows:1. The suitable explant size of Polygonatum stem with bud is 1.0cm×1.0cm×1.0 cm, and the explants are soaked into 0.25%HgCl2 for 18 minutes with the best sterilization effect, the lowest pollution and browning rate, and the highest survival rate.2. The MS culture medium with 3.0 mg/L 6-BA+0.1 mg/L NAA or 3.0 mg/L 6-BA+0.1 mg/L IBA is the best formula for directly inducing rhizome buds from Polygonatum stem with bud.3. In the proliferation culture of Polygonatum rhizome buds, the most suitable medium is MS with 20 g/L or 30 g/L sucrose+3.0 mg/L 6-BA, pH 5.5-6.0 and 12-16 h/d photoperiod. There is no significant difference in proliferation effect among NAA, IBA and IAA. NAA and 6-BA has interacting effects and the best combination is 0.1 mg/L NAA and 3.0 mg/L 6-BA.4. The study has found that the best culture conditions for direct induction and proliferation of Polygonatum rhizome buds is MS culture medium with 0.1 mg/L NAA+3.0 mg/L 6-BA+30 g/L sucrose+7 g/L agar, pH 5.5-6.0 and 16 h/d photoperiod.5.The result shows that 1/2MS culture medium with 2.0 mg/L NAA+2.0 mg/L IBA+5.0 mg/L IAA+0.5 g/L active carbon is the most useful formula for rooting of Polygonatum rhizome buds. A bud develops 20.8 roots with 3.8 cm length after culturing for 9 days, and the rooting rate reaches 100%.6.The best transplanting substrate is a mixture of peat, perlite and sand with a ratio of 1:2:2.
