15 microsatellite makers from Apis mellifera ligustica were used to analyzed the genetic diversity of Local Apis mellifera and Jiuzhou Apis mellifera with each 100 worker bees to study the source material.At the same time,the ten main indexes of external morphology.Gentic variability analyzes within microsatellite DNA polymorphism and the morphology of Local Apis mellifera and Jiuzhou Apis mellifera .The main results were summarized ad follows.1.There existed significant differences(P<0.05) between 10 characteristics .It had significant differences(P<0.05) in length of bee right forewing,width of bee bee right forewing ,length of tergum 3,length of tergum 4 and length of tergum 3+4. Two population had extremely differences(P<0.01) in area of the anterior wing,length of proboscis.It suggested that the Jiuzhou Apis mellifera has more capability of gathering honey.2.Fifteen pairs of microsatellite primers were tested between the two Apis mellifera ligustica ,15 pairs of microsatellite primers can be successfully amplified.The results showed that 86 alleles were found in 15 microsatellite loci,the number of alleles per locus ranged from 3 to 8,mean number of effective alleles was 5.733.The gene heterozygosity(He) of all populations ranged from 0.3145 to 0.7932,and the average value was 0.620±0.138,respectively.The polymorphism information content(PIC) ranged from 0.2829 to 0.7674.,and the average value was 0.593±0.141.All 15 microsatellite loci in this study showed high levels of polymorphism,the reulsts can supply the full information for the analysis heredity multiplicity.3. Fifteen pairs of microsatellite primers were tested between the two Apis mellifera ligustica , The results showed that the everage alleles of Jiuzhou Apis mellifera ligustica was 5.00 Relativly the everage alleles of Local Apis mellifera ligustica was 5.40.The two population's expectation differences are closely,it suggested that the two populations have high heterozygosity and polymorphism, and the high choice potential.4.In this study,the genetic differention between the two population measured as FST value was 4.36%( P<0.01),and measured as FLS value was 0.070(P>0.05), all the loci were contributed to this differention significantly( P<0.01). The genetic differention of the two population, the results showed that the gene between the two population flow greatly.5.The Reynold's distance values of the two population was o.306,and the Nm value was 0.784.The results showed that the Reynold's distance is closely, and Prompts that between two communities possibly to have certain gene exchange. The Local Apis mellifera ligustica comes from Feidong of Anhui,rather than Jiuzhou Apis mellifera ligustica comes from Xuancheng of Anhui,the Reynold's distance of two population is nearly. In addition the honeybee has the strong migration strength, breeding habit and artificial raising (change of scene, business) the influence, cultivates when the strain borrows opposite party blood relationship, the exchange causes these places between the honeybee community to have the certain extent much the gene exchange. Therefore levels of polymorphism between the two population is highly, with this test result match case.
