| The impact of transgenic SCK/CrylAc rice grain on the development of Corcyra cephalonica was investigated by life table methord. The concentrations of CrylAc insecticidal protein in different tissues of the Corcyra cephalonica, feeding on transgenic SCK/CrylAc rice grain were determined by enzyme-linked immunosorbent assay(ELISA). And the activity of enzymes in 3rd instar larvae of the Corcyra cephalonica after being fed on transgenic SCK/CrylAc rice grain was evaluated in the laboratory. The main research results were summarized as follows:1 The result suggested that there were remarkable negative effects on the development of this storage pest. The mortality rate of egg,1 instar larver,2 instar larver and 3 instar larver fed on transgenic rice grain was 100%. And the adults'dry weight of Corcyra cephalonica fed on Bt-transgenic rice grain was remarkable lower than the control. Sex ratio of Corcyra cephalonica fed on transgenic rice grain was also different compared with control.2 The result showed that the levels of CrylAc in Corcyra cephalonica continuously feeding on transgenic rice grain increased with time. Contrariously, the levels of CrylAc in Corcyra cephalonica on control rice grain after feeding on transgenic rice grain for 36 h declined with time. The CrylAc in the pupa, moth, egg were so scarce that it can not been detected by ELISA methord.3 The results showed that the activities ofα-naphthylacetate esterase, alkaline-phosphatase in the larvae fed on transgenic rice grain for 24 h were greatly lower than that of the control fed on the non-transgenic rice grain. However, the carboxylesterase and the acetylcholinesterase activity of the larvae fed on transgenic rice grain increased markedly. The acid-phosphatase activity of the larvae fed on transgenic rice grain for 12 h,48 h and 72 h decreased markedly, being greatly lower than that of the control, respectively. The glutathione-S-transferase in the larvae fed on transgenic rice grain only for 24 h were greatly lower than that of the control. Esterase isoenzymes from both 3rd instar larvae fed on transgenic rice grain for 72 h and that of the control were compared by polyacrylamide gel electrophoresis. The results showed that there were four bands of the esterase in both disposals. The enzymes of the larvae fed on transgenic rice grain were lower active than that of the control, but the removal distance were not changed compared with the control.4 The results showed that the activities of T-AOC in the larvae fed on transgenic rice grain for 12h,36h,48h,72h were greatly lower than that of the control fed on the non-transgenic rice grain. The activities of SOD in the larvae fed on transgenic rice grain for 24h,48h,60h were sharply higher than that of the control fed on the non-transgenic rice grain, however,72h was contray. The activities of POD in the larvae fed on transgenic rice grain for 48h,72h were greatly higher than that of the control fed on the non-transgenic rice grain. The activities of CAT in the larvae fed on transgenic rice grain for 12h,24h,36h,72h were greatly lower than that of the control fed on the non-transgenic rice grain. The content of MDA in the larvae fed on transgenic rice grain was greatly higher than that of the control fed on the non-transgenic rice grain. The activities of resist O2-·activity in the larvae fed on transgenic rice grain for 48h,72h were greatly higher than that of the control fed on the non-transgenic rice grain. The activities of resist OH·activity in the larvae fed on transgenic rice grain for 24h,36h,48h,60h,72h were greatly lower than that of the control fed on the non-transgenic rice grain. |
