Evaluation Of Wheat Leaf Rust Resistance Of 60 Chinese Wheat Cultivars In Chian

Abstract/Summary:
Leaf rust caused by Puccinia triticina is one of the main disease of wheat. Genetic resistance is the most economical and environmental safly measure to control the wheat leaf rust. Knowledge of the leaf rust resistance genes in main planting cultivars(lines) and the genetics in breeding programs were the base for the usage of the resistance cultivars(lines). 60 cultivars collected from Shandong province, Hebei province and Liaoning province were chosen to understand and survey the resistnaceto wheat leaf rust. Clearing the resistance and resistance genes of the test cultivars could provide important information for the rational distribution of production and breeding. The main results are as follows:The DNA extraction method was improved through exploration. Theapproach consists of l00mM Tris-HCl(PH8.0), 1.4M NaCl, 50mM EDTA(PH8.0), 15mg/mL RnaseA form extract dispense with the CTAB, mercaptoethanol,CTAB/NaCl and imitation/isoamyl alcohol (24: 1) . And the method was named as TENR. Experiments showed that the method can be applied to STS, SCAR, SSR marker PCR amplification of high-quality DNA, and TENR method can accelerate the rate of DNA extraction, reduce the pollution and cost and provise the technical support for high-volume extraction of DNA.Sixteen races of P. triticina previously characterized for avirulence/virulence to Lr-genes were uesd to inoculate the test lines in greenhouse for the seedling identification and seedling postulationt. The result of identification was that Shenmian 99042, Shenmian99121, Shenmian99142, Shenmian20162, Shenmian962, Shenmian962, Shenmian85, Shenmian96, Shenmianll67 and Henong5290 showed low resistance to all test races; Henong7106, Shenmian99025 and Shenmian99034 had high resistance to all test races. The result of seedling postulation indicated that the test wheat cultivars may contain wheat leaf rust resistance genes Lr1, Lr2a, Lr3, Lr9, Lrl0, Lr15, Lr16, Lr17, Lr19, Lr23, Lr24, Lr26, Lr29, Lr30, Lr32, Lr36, Lr38, Lr40 and Lr44.Field reactions of the cultivars with test races were determined under field conditions for two years.The result showed that Henong58-3, Henong6251, Henong5290, Weimai8hao, Zemailhao, Laizhou9214, Yan896063, Bin02-47, Weimai7hao, Linmai6hao, Liao9629, Yan5286, Laizhou953, Yan861601, Shenmian99042, Shenmian99121, Shenmian99142, Shenmian20162, Shenmianll67, Shenmian962, Shenmian85 had resistant to leaf rust races with nearly immune reaction. Taishan5024, Liao9629, Zi6135, Taishan269, Weimai6hao, YanC96, Yanl03, Luyuan9160471, Linkang5025, Liao9514 and Shenmian20160 had susceptible reaction. Shenmian99034 and Linmian4hao had moderately susceptible reaction. Shenmian99025 had slow rusting reaction. The result of adult postulation indicated that the test wheat cultivars may contain wheat leaf rust resistance genes Lrl,Lr9,Lrl9,Lr24,Lr38,Lr26,Lr34,Lr36,Lr14b,Lr14ab.In order to determine the resistance genes of the test cultivars, the STS, SSR, SCARE markers closly linked to or cosegregated with Lr1, Lr9, Lr10, Lr19, Lr20, Lr21, Lr24, Lr26, Lr28, Lr29, Lr34, Lr35, Lr37, Lr38 and Lr47 were employed. The results showed that Weimai8, Yannongl5, Zemail, Liao9518, Zi6135, PH01-24, YanC96, Yanl03, Laizhou953, Linmai4hao, Henong6425, Henong9206, Shenmian99042, Shenmian99052, Shenmian99121, Shenmian99142, Shenmian20160, Shenmian20162, Shenmian20167, Shenmian85, Shenmian91 and Shenmian962 may have the leaf resistance gene Lrl site. Shenmian99042, Shenmianll67, Shenmian99121 and Shenmian99142 may have Lr9 site. Zhouyuanl 87 and PH01-35-2 may carry the gene LrlO site. Shenmian20162 may have leaf rust resistance gene Lr24 site, Weimai 8, Zemail, Luhou301, Weimai6hao, YanC96, Yanl03, Yan5158, Yan5286, Binzhou98-6, Luyuan9160471, Yan861601, Henong825, Henong826, Henong6425, Henong827, Henong6251, Henong9206, Shenmian20162 and Shenmian962 may carry Lr26 site. Shenmian99025 and Shenmian99042 may have slow resistance gene Lr34 site. Shenmian20162 and Shenmian962 may contain Lr38 site.F2 families of Shenmian 99142, Shenmian 20162, Henong827, Henong6425, Henong5290,Henong7106were inoculated by the BBB of P. triticina. The result showed that Henong6425 and Henong7106 were controlled by two one dominant genes to BBB. Henong58-3, Henong5290, Henong9914 were controlled by three dominant genes to BBB. Shenmian99142 were controlled by four one dominant genes to BBB. Henong827 were controlled by one one dominant gene.F2 families of Shenmian99142, Shenmian20162, Shenmian99121, Henong5290, Henong826, Henong827, Henong6425 were inoculated by the others of P. triticina. Base on the genetic analysi and moleculer marker-assisted identification, the resistane of Shenmian99121and Shenmian99142 to FCGT was supposed controlled by Lr9, and Shenmian20162 was confered by Lr24. Henong6425 to FBQT was supposed controlled by Lrl and Lr26. Henong827 and Henong826 to FBQT were supposed controlled by Lr26. The resistance of Henong5290 was controlled by one semi-dominant unknown gene at seedling stage, and by one unknown dominant gene at adult stage.Henong58-3, Henong6251, Henong5290, Weimai8, Zemail, Laizhou9214, Yan896063, Bin02-47, Weimai7, Linmai6, Yan5158, Liao9629, Yan5286, Laizhou953, Yan861601, Shenmian99042, Shenmian99121, Shenmian99142, Shenmian20162, Shenmian 1167, Shenmian962 and Shenmian85 have good potentical in wheat leaf rust resistance breeding. Taishan5024, Liao9629, Zi6135, Taishan269, Linkang5025, Liao9514, Weimai6hao, YanC96, Yanl03, Luyuan9160471, Linmai4hao, Henong827, Shenmian 20160 were not recommened to use and promote in the wheat leaf rust-prone areas.
