The results as follows:Isolating Rhizobium from the root nodule which collected from Shandong province, and getting 89 Rhizobium strains by Back inoculation experiments. Divided Isolated Rhizobium into five groups through the IGS and BOXAIR typing method; The results of the matching experimental of five groups selected from strains randomLy with soybean varieties showed that it is Significant difference of Nodulation and Nitrogen Fixation between different Rhizobium strains of soybean. It showed more relatively strict matching of fast-growing soybean rhizobia root nodule than the slower-growing soybean varieties showed; The USDA110,USDA205,Shanxi4-1,M-5,JJ-5-4 strains had a more broad-spectrum of the symbiotic nodulation; In the Matching Integrated, the strain M-5 can participate in experiments with all varieties of soybean nodule, and In addition to hedou 12, the ability of nodulation of strain HH-1-6 with other species is unobvious or not. In the soil culture experiment, M-5 strain showed good nitrogen fixation ability; These two Rhizobium strains is an ideal material to research the mechanism of Rhizobium and host symbiotic nodulation. Two strains were identified to Sinorhizobium by 16S rDNA analysis.The protein expression profiles of M-5,HH-1-6 were analyzed after they were disposed to daidzein of hedou 12.The results showed that:M-5(Sinorhizobium) had 23 up-regulated proteins and 1down-regulated proteins in expression with significant difference,while HH-1-6(Sinorhizobium) had 22 up-regulated and 1 down-regulated proteins. Differentially displayed proteins were analyzed by MALDI-TOF-MS, and 9 of them were identified from M-5 and 4 proteins from strain HH-1-6.In this research, two signal transduction proteins PfkB/PBP1 and a series of rhizobia with growth and nodulation related proteins were founded in prominent expression protein in strain M-5 (Sinorhizobium) and four related with growth of energy metabolism and close relationship in strain HH-1-6 (Sinorhizobium) after exposed to soybean daidzein. Two strains all showed obvious response from genetic level , but how to determine thekey genes that effects of nodulation matching are further study. |
