Pinus tabulaeformis f.Shekannesis woods are only located in Ziwuling Loess Plateau (Shaanxi Province) 337.3 hectares totally. Comparing with pinus tabulaeformis, Pinus tabulaeformis f. Shekannesis has many advantages, known as the precious genetic resources in the loess plateau, it is very important to protect this species. In this research, genetic diversity of two extant populations of Pinus tabulaeformis f. Shekannesis were investigated by using RAPD markers. The results were showed as follows:(1)The fact that DNA from the shoots of pinus-arten can be used for RAPD expand had been confined positively.(2)50 random primers selected from 500 random primers had polymorphism and totally 349 loci were generated.Thereinto,321 loci had polymorphism which occupied 92.51%.Each primer could augment 4-14 loci with 7 loci in the average. The polymorphism information content among the 50 primers were analyzed, S3,S8,S11,S19,S29,S40,S46,S50,S52,S102,S237, S249,S293,S334,S358 and S404,got the highest PPB 100﹪, S32, S290, S323 and S375 got the lowest 70﹪,with the average 89.92% among the 50 primers.(3)The results showed that Pinus tabulaeformis f.Shekannesis have an extraordinarily high level of genetic diversity. on the species level, percentage of polymorphic loci (PPB) was 95.51%, total gene diversity(Ht)was 0.3174, Nei's gene diversity (H) and Shannon's Information index (I) were 0.3173 and 0.4566 respectively. on the population level, mean percentage of polymorphic loci was 0.8992%, the means of H and I were 0.3025 and 0.4566 respectively.(4)A low level of genetic differentiation between populations was detected based on gene flow(Nm =10.2018), Nei's genetic diversity analysis (4.67%) and Shannon's diversity index (4.52%).(5)Finally, Based on the results mentioned above,some management strategies of protecting this species were proposed.
