Pinus tabulaeformis Carr. is the most important conifer species in northern China. It has function of good landscaping and soil and water conservation, powerful ecologal function, it has tremendous value in the timber industry and pine resin,pharmaceutical industry. Random amplified polymorphic DNA (RAPD) markers were used to measure genetic diversity of 192 individuals from 12 natural populations(Shannxi,Neimeng,Gansu,Shanxi province) of Pinus tabulaeformis Carr.The main results are as follows:(1) Established a effective method to extract the genome DNA from the sprout of Pinus tabulaeformis Carr. poplar for RAPD.(2) 20 random primers selected from 127 random primers had polymorphism to 192 Pinus tabulaeformis Carr. DNA, and A total of 289 loci were identified, of which 220 polymorphic bands, the total number of amplified bands 76.12 %. Primers S46 and 519 the highest percentage of polymorphic bands amplified, to 100.00%; while the percentage of polymorphic bands primer S344 lowest, only 46.67%.(3) Pinus tabulaeformis Carr. has a high level of genetic diversity. At the species level, PPL was 76.12%, H was 0.3157, I was 0.4683. Within the population, the average percentage of polymorphic loci was 54.03%, the average of H, and I were 0.2545 and 0.3848 respectively.(4) The genetic differentiation among Pinus tabulaeformis Carr. populations is higher. Gst was 0.1939 ,Nm was 2.0786, (Isp-Ipop) / Isp is 0.1783. The result show that the genetic variation of Pinus tabulaeformis Carr. exist within populations, more than 80% of the total variation, and the genetic variation among populations accounted is about 20% of the total variation.(5) Established the genetic similarity coefficients by UPGMA (un-weighted pair group method with arithmetic) analysis. And on level 0.76, the result showed that the 12 natural populations could be clustered into four groups.(6) Some management strategies and protect measures are proposed for conservation planting.
