It has been studied about that the solid fermentation culture of Wild Phellinus, species identification and phylogenetic evolution study of wild Phellinus based on rDNA ITS sequence analysis in this research.Since wild Phellinus issubjected multiple constraints of the external environment, the formation of fruiting bodies in nature than the rare, but artificial cultivation of environmental and technical constraints are another big plus, Phellinus increasing the amount of another, especially South Korea, Japan, wild resources in China predatory acquisition, Phellinus there has been less and less volume. Fermentation using the Phellinus mycelium, which contains the ingredients are similar to the fruiting body of the active ingredient itself, but also relative to the artificial cultivation of short cycle time, in the near future a large number of Phellinus no mycelium subject to external constraints, significant reduction in the medicinal fungus as a"medicinal"cost, yield and quality are relatively high, for further study, Phellinus laid the foundation of mycelium, using fermentation, Phellinus mycelium increasingly affected by human concerns. Therefore, wild Phellinus issubjected multiple constraints of the external environment to get through artificial methods of cultivation, Phellinus fruiting body and the materials needed for later molecular identification. Research shows that separated activating Phellinus mother culture of fruiting body tissue isolation, with the common potato dextrose agar (PDA) medium, as pH6.0, temperature 28℃, 55% water content, the most appropriate environment for mycelium body growth. Phellinus strains for the production of mycelium and fruiting body cultivated fermentation medium and conditions for a formula to provide a reference for further study of species identification and phylogenetic evolution study of wild Phellinus.Currently, the Phellinus species name are disputed, the main from the fruiting bodies of fungi, because of the identification difficult, making medicinal fungi, Phellinus in the development and application,frequently"synonym","homonym"phenomenon.Morphology of the initial judgments can only be symbolic, not a fundamental identification Phellinus wild strains 'species' taxon, and the final classification and identification of fungi depends on the determination of sequence analysis were tested strains with the gene sequences homology with known fungi. This allows a small number of sequencing errors or a small amount of mismatch, not only for classification and identification, but also on the fungal taxa for phylogenetic analysis. rDNA ITS sequence analysis using the two universal primers (ITS1 and ITS4), PCR amplified fragments of 10 entries, will be tapping PCR amplification and purification, cloning and sequencing, sequencing by MEGA4.1 software results and GenBank, similar to the known sequence of the cluster analysis, using Neighbor-Joining phylogenetic tree was constructed. Experimental results show that bacteria are used in the genus Phellinus, that Phellinus. Where S1, S4, S6, Rh, Sc identified as Powell Phellinidium that Phellinus baumii; S2, S3, S5, S7, Gy identified as Cleftho of Phellinidium that Phellinus linteus.
