Phellinus linteus has the effect of blood circulation,hemostasis,clearing liver and relieve turbity formula and antidiarrheal.Currently,as the large demand of market,the lack of resources of wild Phellinus linteus,and the difficulty of artificial cultivation.So,it has great significance to obtain a large amount of mycelium and active substances by solid and liquid fermentation.This paper studies phellinusigniarius from four aspects:First,study on the optimization of experimental culture medium for phellinuslinteus solid and liquid;Second,artificial identification of mulberry Phellinus,including identification of biological characteristics,macroscopic observation of fruiting body morphology,microscopic observation of microscope and scanning mirror,and molecular identification base on ITS sequence;Third,identifie the Phellinus linteus which collected from different species,compare with their appearance,polysaccharides,flavonoids and other active ingredients.Fourth,compare the contents of different species of Phellinus linteus fruiting body of trace elements Ca,Mg,Fe,Zn,Cu,Se,Pb,K.The following results are obtained.The optimum nitrogen source for solid culture is the integrated nitrogen source of soybean and Maize,the optimum carbon source is glucose,the optimal initial pH is 6,the optimum culture temperature is 27-31.The indexes of biomass,extracellular polysaccharide and extracellular flavonoids determined the optimum conditions of fermentation of Phellinus igniarius:soybean + corn is 100g/L,glucose is 30g/L,KH2PO4 is 3g/L,MgSO4 is 0.75g/L,vitamin B1 is lmg/L,the initial pH is 6.Liquid culture changes during the process of extracellular enzymes that:Phellinus has abundant extracellular enzymes,the peak of amylase activity appeared in the first.The peak activity of cellulose degrading enzyme include cellulose degrading enzyme filter paper cellulose and carboxymethyl cellulose appeared in the first 6d,The activity peak of laccase enzyme include laccase,guaiacol oxidase and polyphenol oxidase respectively appeared at 8d,10d,10d,The change of enzyme showed that the utilization of external nutrients during the growth of mycelium.The content of reducing sugar decreased gradually with the culture time,and there were asmall peaks at 6d and 8d.The changes of Protein concentration and extracellular enzyme activity substantially in the same,there were appeared peak in the 6d and 8d respectively.The wild Phellinus fruiting bodies can be parasitic and rot on different broad-leaved trees.Mulberry fruiting bodies of Mulberry,Poplar,Lilac tree,Pine were collected,Through the study of the biological characteristics,and the observation of appearance description to identify the authenticity of Phellinus,Through the molecular identification of ITS sequence,further determined the different species of Phellinus species,The results show that the Phellinus of mulberry belongs to Phellinus rhabarbarinus;Phellinus of clove tree belongs to Phellinus Inonotusbaumii;poplar of Phellinus belongs to Fuscoporiagilva;Phellinus of pine belongs to Phellinus hartigii.Through the comparative analysis of the active ingredient which were the Polysaccharide and flavone content of Phellinus fruiting bodies,Preliminary judgmented the quality of active ingredients of mulberry Phellinus,antitumor polysaccharide content was significantly higher than that of other species of fruiting.The trace elements Ca,Mg,Fe,Zn,Cu,Se,Pb,K were compared between the mycelium and the fruiting bodies of different sources,The content of the phellinusigniarius Fe,Zn,K higher than the fruiting body and the harmful heavy metal Pb is lower than the fruiting body.
