In this study, using the classical genetic method and SSR markers to analysis of resistance to stripe rust in three villosa translocation line V9125-2, V9125-3 and V9125-4 at seedling stage.Then,determined and mapping the location of resistance gene to stripe rust.The genetic analysis of three Triticum aestivum-Haynaldia villosa translocation lines,V9125-2,V9125-3 and V9125-4,were investigated.The results are as follows:1.Using seven prevalent races of yellow rust CYR29,CYR30,CYR31,CYR32, CYR 33 and Sun11-4,Sun11-4, Triticum aestivum-Haynaldia villosa,7182 and the three Triticum aestivum-Haynaldia villosa translocation lines,V9125-2,V9125-3 and V9125-4,were evaluated to stripe rust repeatesly.The results showed that the three Triticum aestivum-Haynaldia villosa translocation lines were high resistance to the current epidemic yellow rust races.2. Genetic analysis of F2 hybrided of three wheat-Haynaldia villosa translocation lines resistance to stripe rust, the results showed:(1) One dominant gene conferred resistance to stripe rust race CYR29,one dominant gene and one recessive gene indenpently conferred to CYR30,CYR32,CYR 33 and Sun11-11,and two pairs of independent dominant genes conferred to CYR31 and Sun11-4 in V9125-2.(2)Two pairs of independent or overlap genes conferred to CYR29,CYR30 and CYR31, one dominant gene and one recessive gene indenpently or overlap conferred to CYR32,CYR33,Sun11-11,and two pairs of complementary genes conferred to Sun11-4 in V9125-3,(3)Two pairs of complementary genes conferred to CYR29 and CYR31,two pairs of independent or overlap genes conferred to CYR30,Sun11-4 and Sun11-11,and one dominant gene and one recessive gene indenpently or overlap conferred to CYR32 and CYR33 in V9125-4.3. The F2 population innoculated with CYR29 were used for SSR analysis, temporarily designated as YrWV.SSR markers, included 289 pair primers,combined with bulked segregation analysis (BSA) revealed that six SSR markers, Xbarc87, Xwmc463,Xwmc405,Xbarc126,Xwmc438 and Xgwm473, located on chromosome 7DS were linked to YrWV, and their genetic distances to YrWV were 9.1,3.9,5.1,12.6,29.0 and 57.4cM, respectively,the six sites linked to YrWV were validated by a segregated ratio of F3 lines.These markers would be used for breeding new wheat cultivars with marker-assisted selection. It is very low homologous rate for 43 varieties in Huang-Huai Wheat grown region tested with YrWV.It is expounded inheritance and location of the resistance gene to stripe rust of the three wheat-Haynaldia villosa translocation lines. The reseach provide a basis of exploid and utilize the new genes in wheat stripe rust resistant molecular marker-assisted breeding.
