The potato non-viral production system established by means of tissue culture is a kind of very effective high-tech biotechnology, which can greatly improve yield and quality of potatoes. This experiment takes Japanese potato (Solanum tuberosum L.) leaves as the test materials, which have high starch content, drought tolerance and the strong root system. Through the selection of the best reproductive system in the stage, the optimum ratio of plant hormone concentration is obtained. During the process of the test, the observations over biological characteristics of tissue culture seedlings of potatoes provide theoretical basis on potato rapid reproduction and the theories. At the same time, the way that in vitro rapid propagation of potatoes uses tissue culture of rapid propagation techniques, not only has directive significance and broad prospect of application in the production process, but also has important meanings of improving its quality and yield with mass production of potato detoxification. The results of experiments were as follows:(1)The optimal media for Janpanese potato induction from callus was MS + 6-BA 2.5 mg·L-1 + NAA 0.3 mg·L-1.(2)The optimal media for subculture callus was MS + 6-BA 2.0 mg·L-1 + NAA 0.3mg·L-1 + GA3 10 mg·L-1.(3)The optimal medium for adventitious bud differentiation from leaf-derived callus was MS + 6-BA 2.5 mg·L-1 + IAA 0.1mg·L-1 + GA3 1.0 mg·L-1.(4)The optimal medium for adventitious root induction from callus was MS + NAA 1.0 mg·L-1.
