The experiment conducted to study the effect of milk protein precursors concentration variation of external pudic artery on mammary gland intake and related hormone receptor mRNA expression of mammary gland tissue in milk goat. Six 2-3 years old milk goats were chose, which have nearly body weight and milk yield, with the vessel intubation of external pudic artery and milk vein. Six milk goat is six repeats, every milk goat perfused essential amino acid that have different deletion quantity, no perfused is control group. Used arteriovenous concentration difference technique to research the effect of perfused 3h(4.65g 75% deletion quantity), 4h(6.2g 100% deletion quantity) and 5h(7.75g 125% deletion quantity) on mammary gland intaked milk components precursors, used molecular technique to research the effect of milk protein precursors concentration variation on related hormone receptor mRNA expression of mammary gland tissue.(1) Blood flow of six milk goat were measured, the results showed that the average mammary gland blood flow is 124.76L/d, the average mammary gland plasmaflux is 83.17L/d. Calculated the ratio of mammary gland blood flow and milk yield, the results fit 500: 1 in principle.(2) Perfused essential amino acid increased the intakes of EAA and NEAA. After perfused essential amino acid, the mammary gland EAA intake of treatment groups were higher than control group significantly(P<0.05), and there were no significantly difference among each treanment group(P>0.05). The intakes of Thr, Met, Lys were all higher than control group(P<0.01). Compared with control group, the intakes of Ser and Pro were not significantly(P>0.05), other non-essential amino acid intakes of treatment groups were higher than control group(P<0.05). The NEAA intakes of treatment groups were higher than control group very significantly(P<0.01). Compared with control group, the glucose, TG and TC contents have significant difference(P<0.05), the urea nitrogen has no difference(P>0.05). Increased with the priming volume, the contents of milk protein and lactose were step up(P<0.05), the contents of milk fat and solids-not-fat were step up but not significantly(P>0.05).(3) The blood plasma IGF-I, GH and PRL of three treatment groups were all higher than control group(P<0.05), the best effect in treatment groups is perfused 100% deletion quantity of EAA. The real time fluorescent quantitation PCR found that after perfused EAA, the expression of three hormone receptors mRNA in mammary gland tissue, trentment groups were higher than control group very significantly(P<0.01).
