Nowadays, the management of strains of edible fungi is in chaos.The phenomeon of different species with one name and one species with different names is quite serious.But we still do not have a standard strain library which can be used to classify strains of edible fungi and identify new strains quickly.Therefore,we establish standard strain library of Pleurotus eryngii, Pleurotus nebrodensis and Pleurotus ferulae respectively based on strains stored by Microbiological Research Center of Fujian Agriculture and Forest University,in which every strain is totally different from the other.The main content and results of the research are as follows:1. The establishment of standard strain library of Pleurotus eryngii, Pleurotus nebrodensis and Pleurotus ferulaeStandard strain library of Pleurotus eryngii, Pleurotus nebrodensis and Pleurotus ferulae were established based on 27 strains of Pleurotus eryngii,20 strains of Pleurotus nebrodensis and 8 strains of Pleurotus ferulae respcetively with antagonistic reaction and SCAR. Standard strain library of Pleurotus eryngii includes 26 strains, standard strain library of Pleurotus nebrodensis incudes 19 strains,while standard strain library of Pleurotus ferulae includes 8 strains.2. The complement of the information of standard strain library of Pleurotus eryngii, Pleurotus nebrodensis and Pleurotus ferulaeThe hyphae biological characters, agronomic characters, polarity of some strains of standard strain library were researched. The hyphae biological characters were researched in three aspects,which wre colony morphology,hyphae growth rates and the effect of different temperatures,humidity and pH values on hyphae growth rates.The results showed that the hyphae biological characters of strains in standard library are different from each other. Agronomic characters were researched in six aspects,which were hyphae growth rates,yields,the diameters of pileus and stipe,the length of stipe and the thickness of pileus. The results also showed that Agronomic characters of strains in standard library are different.The polarity of 11 strains of Pleurotus eryngii and 9 strains of Pleurotus nebrodensis were tested.The result showed that Pleurotus eryngii has a abundance of multiple alleles(A1B1- A7B9),while Pleurotus nebrodensis has a few multiple alleles(A1B1- A5B5).3.The implement of standard strain library of Pleurotus eryngiiThe rest strains of Pleurotus eryngii were classified by SCAR and antagonistic reaction,and then intercompared with strains in standard strain library that had been established.The result showed that there are 13 strains that has the phenomeon of different species with one name,which takes 18.06 percent of all strains of Pleurotus eryngii stored in our Lab.4. Spawn Preservation and enzyme activity assayOne strain of Pleurotus eryngii, Pleurotus nebrodensis and Pleurotus ferulae was preserved with four methods(liquid nitrogen preservation,distilled water preservation,paraffin preservation, 25℃preservation),germination rate, germination time and hyphae growth rate after four methods of preservation was measured and the enzyme activity of carboxymethy cellulase,laccase,amylase were compared.Liquid nitrogen preservation is the best method.And then,four factors influencing the effect of liquid nitrogen preservation were optimized.The result showed that using young strain, saw dust as culture media, 10% glycerol as antifreeze, cooling rate of 1℃/ min from 4℃to -40℃as the cooling process is ideal.
