| In this research, an identification method was established to identify Volvariella volvacea by DNA molecular markers techniques such as RAPD and ISSR, observation of antagonism and colony morphology, and 30 standard strains have been found. Further identify and verify of the 30 standard strains were used by 11 SCAR markers from Mycological Research Center of Fujian Agriculture and Forestry University. Then three strains were homonym. Finally, a standard strains bank of Volvariella volvacea containing 28 standard strains was built.The hyphae biological characters and agronomic characters of 28 standard strains were researched. To investigate how the 28 standard strains grew on different temperature, pH value, moisture of cultivation material and growth rate in test tube. Many parameters of agronomic characteristics such as product yield had been conducted by fruiting test. An evaluation of the 28 standard strains had been drawn from those data, which provided information for breeding and cultivating of Volvariella volvacea.32 strains were preliminarily divided into 21 groups by the established fast identification system in this research. Those 21 groups were further divided into 26 groups by antibiosis test. Finally, antibiosis test was conducted between the 32 strains and the 28 standard strains, and all the 62 strains conserved in MRC were divided into 50 groups.In addition, the method of spawn preservation of Volvariella volvacea was discussed. The results showed that the best method is liquid nitrogen preservation. At the same time, the conditions of liquid nitrogen preservation were optimized. The optimum conditions: PDA medium, 10% glycerin antifreeze, program 9 ( 25℃~ 4℃using 9℃/min cooling rate; 4℃~–40℃adopt 1℃/min cooling rate; after reduce temperature directly to–196℃) cooling and 35℃thawing activation.
