Lilyturf is general designation of a part of Liriope and Ophiopogon genus in liliaceae. Lilyturfs have high value in medicinal and greening.In this paper germplasm resources of lilyturf were investigated and collected, a part of them were studied on adaptability of introducing. It was the first time both at home and abroad that SRAP markers had been used in genetic diversity of 48 lilyturf belonged to about 20 species. The main results are as follows:Systematic survey on the plants with the name of lilyturf both at home and abroad had been carried on. The textual research for The latin names, English names, Chinese names of 38 species or varieties of lilyturfs were nvestigated. Investigation showed that foreign countries had abundant cultivars, main of which had high ornamental value. They were widely used in The United States, Japan and Korea, etc.. Through field investigation and other methods,196 germplasms about were collected from 13 provinces, Yunnan, Sichuan, Guangxi, Guangdong, Zhejiang, Shanghai, Jiangxi, Hubei, Fujian, Jiangsu, Anhui, Henan, and Beijing. They were identified as 25 species or varieties. The germplasm preservation conditions and cultivation measures of lilyturfs were preliminarily studied.The adaptation of more than 20 liliturfs were studied in Beijing for two years, and their phenophase and other biological characteristics, conditions of over summering and overwintering, shade, drought and heat resistance, occurrence condition of diseases and insect pests were studied. Cold hardiness was focused on study,8 physiological indicators of 11 liliturfs were measured under cold stress. Result showed, Low temperature was a key factor for introducing lilyturf to Beijing. Only a few species or varieties, such as Ophiopogon japonicus, Liriope spicata, Liriope muscari, Liriope platyphylla, Ophiopogon intermedius cv. Argenteo-marginatus, Liriope muscari var. variegate, could overwinter and adapt Beijing climate.High quality genomic DNA extraction methods of lilyturfs were established in this paper. By the improved CTAB method and the improved SDS method, high quality genomic DNA was separately extracted from leaves of lilyturfs included in recalcitrant plants. the improved CTAB method had high DNA purity, good generality, it could meet a variety of molecular biology experiments.A rapid and efficient polyacrylamide gel electrophoresis and silver staining method was established. Compared with conventional methods, the total time of making gelatin, electrophoresis, silver staining process shortened by about 3 hours, it was good for large-volume primers screening. Silver staining established in this study had a clear staining background, saving reagents, and easy to operate. 48 lilyturf germplasms were detected on the genetic diversity used 11 primer pairs screened from 64 primer pairs. A total of 621 bands were produced, all were polymorphic bands, with polymorphism rate of 100%. Every primer pair produced 56 polymorphic bands on average.Cluster analysis showed that the Liriope and Ophiopogon species had no obvious boundaries, supporting the view that the two genus should be classified as one genus. Ophiopogon japonicus and Liriope spicata had great genetic diversity within species. Liriope muscari and Liriope platyphylla had close genetic distance each other, so considering Liriope platyphylla as a short variety of Liriope muscari was reasonable.
