In this paper, the binding interaction between colchicine and protein of ginkgo male strobilus was studied by ultmviolet absorption and fluorescence spectra;Extractions of colchicine from the protein of different parts of ginkgo by organic solvent were studied respectively, and the colchicine content was determined by high performance liquid chromatography (HPLC), identified binding force between colchicine and proteins in different tissues of ginkgo. This study applied proteome method to study the proteins expression pattern of ginkgo male strobilus under colchicine treatment. From the physiological and biochemical molecular level reveal specificity that colchicine induced ginkgo chromosome doubling.To investigate of mechanism ginkgo microspore doubleGinkgo male strobilus protein absorption peak near 280nm and 295 nm with colchicine concentrations increased and blue shift occurred to some extent, the processing time have almost no effect in a system; 278nm and 295nm excitation, the colchicine were caused ginkgo male strobilus decreased protein intrinsic fluorescence, fluorescence emission peak position have occurred in the degree of blue shift; colchicine treatment of protein of ginkgo male strobilus, with the temperature, the fluorescence intensity.High Performance Liquid Chromatography determine the amount of free colchicine. Identified the binding force between colchicine and protein of ginkgo. Ginkgo male strobilus protein binding colchicine and activity is the strongest one for 7.977 ng.mg-1.ml-1, followed by Ginkgo embryo protein, as 2.701ng.mg-1.ml-1, ginkgo leaf protein weakest, to 2.519 ng.mg-1.ml-1 Optimized technology of Ginkgo male strobilus protein 2-DE. The results showed that the spots in protein map in TCA/acetone were more than that in phenol and in Tris-Hcl.The protein loading of 1500ug could obtain clear map with more protein spots and better separation effect.This study use 2-DE gel electrophoresis to study the proteins expression pattern of ginkgo male strobilus under colchicine treatment. By comparison, six different protein spots was found, two proteins were emerging, one protein spots disappeared, the expression of a protein spots up and down two proteins.
